Cryo-electron microscopy is addressed using cryo-EM to examine archaeal RNAPs from Euryarchaeota and Crenarchaeota alongside yeast RNAP II and suggests that evolutionary tuning of clamp flexibility and dynamics contributes to distinct GTF-dependent promoter-loading mechanisms.
