Sigma-1 receptor activation mediates a faster antidepressant-like effect through disinhibition of the DRN5-HT-mPFCpyr neural circuit via S1R/5-HT1AR heterocomplex.

BACKGROUND The Sigma-1 receptor (S1R) represents a promising target for faster-onset antidepressants. Nevertheless, the mechanism underlying the delayed onset of therapeutic action exhibited by S1R-activating selective serotonin reuptake inhibitors (SSRIs) remains unclear. Thus, elucidating the role of S1R in mediating...

GPCR heteromers: Beyond the monomeric paradigm in neuropsychiatric drug discovery

G protein-coupled receptors (GPCRs) constitute the largest druggable superfamily of membrane proteins and underlie approximately 35% of all approved medicines. Yet this conventional paradigm leaves a substantial minority of neuropsychiatric patients with incomplete efficacy, treatment resistance, or dose-limiting adver...

Activation of sigma non-opioid intracellular receptor 1 (S1R) receptor inhibits plexin domain containing 2 (Plxdc2) driven microglial inflammation to ameliorate depressive-like behaviours in mice.

Analysis demonstrated that overexpressing Plxdc2 in mPFC microglia activated the JAK2-STAT1 pathway, enhanced NLG, shifting microglia towards a pro-inflammatory phenotype, worsening deficits in synaptic plasticity and induced depressive-like behaviour, which was reversible by the NLG inhibitor tunicamycin.
