CircACVR2A(2–4) regulates stem cell pluripotency by uncoupling CDK-activating kinase functions

Abstract Differentiation of stem cells is driven by tightly coordinated transcriptional programs, yet the contribution of circular RNAs (circRNAs) to lineage specification remains poorly defined. Here, we profiled circRNA and messenger RNA expression during differentiation of human embryonic stem cells into five germ l...

A human iPSC-based perfusable and membrane-free neurovascular unit-on-chip connecting brain organoids to the blood–brain barrier

The blood–brain barrier (BBB) protects the central nervous system by restricting entry of harmful blood-borne factors, but this selectivity actively limits delivery of therapeutics to the brain. Because BBB function is shaped by dynamic interactions within the neurovascular unit (NVU), particularly between brain endoth...

Single-neuron resolution mapping of dopaminergic connectivity across development, adulthood, and degeneration.

This work developed and employed a unique intersectional genetic strategy in mice that enables specific sparse labeling of mDA neurons across the midbrain from early developmental stages onward and generated the largest dataset to date of 3D-reconstructed mDA neurons at key developmental stages, in adulthood, and in mo...

Pathology-defined cell states reveal reproducible transcriptomic signatures across ALS cortical single-nucleus RNA-seq studies

It is demonstrated that pathology-defined cell states provide a more reproducible framework for studying ALS transcriptomic alterations than conventional case-control comparisons and an interactive browser is provided to facilitate exploration and comparison of ALS snRNA-seq datasets.
