724. Clinical utility of novel antipsychotics in schizophrenia

Abstract Background Antipsychotic development in schizophrenia has been constrained by reliance on dopamine D2 receptor blockade, yielding modest incremental efficacy alongside persistent adverse-effect burden. Recent agents and late-stage pipelines aim to expand mechanisms of action, most notably via central muscarini...

Relapse trajectories and antipsychotic discontinuation in schizophrenia from individual participant data of five trials from the Yale Open Data Access database: a meta-analysis.

It is found that rapid relapse was not over-represented among those who discontinued treatment, which is not the pattern expected if rapid relapse following paliperidone discontinuation were commonly a pharmacological discontinuation effect.

Striatal dopaminergic function and cortical functional connectivity in 22q11.2 deletion syndrome: an integrative multimodal 18F-DOPA PET and magnetic resonance imaging study.

Dopaminergic signalling is critical for regulating large-scale brain network dynamics and is implicated in the pathophysiology of psychotic disorders. 22q11.2 deletion syndrome (22q11DS), a high-penetrance genetic risk factor for schizophrenia, is associated with both dopaminergic alterations and disruptions in functio...
