Treatment of multiple sclerosis with peptide-coupled red blood cells induces antigen-specific T regulatory cells.

Inducing antigen-specific immune tolerance is a long-standing aim in organ-specific autoimmune diseases including multiple sclerosis (MS). We developed a therapy based on autologous red blood cells (RBCs) as tolerizing carriers of autoantigens and tested these in an open-label, dose-escalation, phase Ib clinical trial...

DNA methylation landscape of cerebrospinal fluid cells in multiple sclerosis: an epigenome-wide association study

MS-associated methylation changes enriched in the cohesin chromatin-regulation pathway localised to T-cell regulatory regions included multiple protocadherin (PCDH) genes, which displayed consistent methylation and expression changes in CSF cells of pwMS compared to controls.

Genome-wide association analyses highlight the neuronal contribution to multiple sclerosis susceptibility

Inhibitory neurons emerged as a key target cell type for MS-associated variants, with seven loci, including STAT3, displaying altered expression only in these cells, suggesting that MS susceptibility may reflect reduced central nervous system resilience to inflammatory challenges.
