Novel Lomefloxacin–Triazole Hybrids: Design, Synthesis and In Vitro Antiproliferative Activity in Breast, Melanoma and Colon Cancer Cell Lines

Promising antiproliferative activity of the tested derivatives of lomefloxacin with a 1,4-disubstituted-1,2,3-triazole moiety is revealed, particularly against colorectal cancer cell lines.

Mechanistic insights into EB171 anticancer activity in MCF7 and MDA-MB-231 breast cancer cells showing differential responses.

Mechanistically, EB171 induced replication stress and DNA double-strand breaks, accompanied by activation of cell cycle checkpoints, cyclin suppression, and p21Waf1/Cip1 upregulation, and these findings support further investigation of EB171 as a promising anticancer compound.
