Parasedimentitalea nitratireducens sp. nov., a Novel Nitrate-Reducing Bacterium Isolated from the Yellow Sea

A Gram-stain-negative, rod-shaped, aerobic strain CY06T was isolated from surface marine sediment sample of the Yellow Sea, China. The strain was positive for oxidase and catalase and could reduce nitrate to nitrite but not further to gaseous nitrogen products. Phylogenetic analysis based on 16S rRNA gene sequences and the up-to-date bacterial core gene (UBCG) sequences revealed that strain CY06T formed a distinct clade within the genus Parasedimentitalea. Genomic comparisons, including digital DNA-DNA hybridization (dDDH), average nucleotide identity (ANI) and average amino acid identity (AAI), clearly distinguished strain CY06T from other members of the genus. The morphological, physiological, biochemical and chemotaxonomic data further supported its differentiation from closely related species. Based on the polyphasic evidence, a new species, Parasedimentitalea nitratireducens sp. nov., is proposed, with the type strain CY06T (=MCCC 1K08636T = KCTC 72006T).

Hyunsoonleella zeaxanthinifaciens sp. nov., Isolated from Coastal Seawater of Jeju Island, Republic of Korea

A Gram-stain-negative, strictly aerobic, yellow-pigmented, non-motile, rod-shaped rod-shaped bacterium, designated J2K-J805T, was isolated from coastal seawater of Jeju Island, Republic of Korea. Strain J2K-J805T grew at 15–37°C (optimum, 30°C), pH 6.0–8.0 (optimum, pH 7.0–7.5) and in the presence of 1–4% (w/v) NaCl (optimum, 2–3%) and was catalase-positive and oxidase-negative. The strain showed the highest 16S rRNA gene sequence similarity to Hyunsoonleella flava T58T (98.4%). Phylogenetic and phylogenomic analyses based on the 16S rRNA gene and whole-genome sequences placed strain J2K-J805T within the genus Hyunsoonleella as a distinct lineage. The OrthoANIu and digital DNA-DNA hybridization values between strain J2K-J805T and H. flava T58T were 87.7 and 34.5%, respectively, both below their respective thresholds for species delineation. The genome size and DNA G+C content were 3,825,400 bp and 34.9 mol%, respectively. The predominant respiratory quinone was MK-6, and the major cellular fatty acids were iso-C15:1 G, iso-C15:0 and iso-C17:0 3-OH. The polar lipid profile comprised phosphatidylethanolamine, three unidentified aminolipids, two unidentified glycolipids and five unidentified lipids. High-performance liquid chromatography with photodiode-array detection identified the major yellow pigment as zeaxanthin, and the genome contained the carotenoid biosynthesis genes crtB, crtI, crtY and crtZ together with genes for isoprenoid precursor supply. On the basis of these polyphasic data, strain J2K-J805T represents a novel species, for which the name Hyunsoonleella zeaxanthinifaciens sp. nov. is proposed. The type strain is J2K-J805T (= KCTC 102498T = JCM 38469T).

Haloleucobacter ellipsoideus gen. nov., sp. nov., and Haloleucobacter albus sp. nov., two novel N2-fixing hydrogen bacteria isolated from soil near a wastewater treatment plant.

Two Gram-stain-negative, aerobic, oxidase-negative, non-motile, ellipsoid to short rod-shaped bacteria (~1.2-2.0 µm in length and 0.7-0.8 µm in width) were isolated from soil near a wastewater treatment plant in Hangzhou, Zhejiang Province, China, designated TB0136T and TB0139T. Both strains are able to grow chemolithoautotrophically using hydrogen as an electron donor and perform nitrogen fixation in the absence of ammonium or any organic nitrogen source. Strains TB0136T and TB0139T were observed to grow at temperatures of 15-45 °C (both optimal at 33 °C; TB0139T additionally showed trace growth at 50 °C), at pH of 6.0-10.0 (TB0136T optimal at 7.5-8.0, TB0139T optimal at 8.0-8.5) and in the presence of 0-6.0% (w/v) NaCl (optimal at 1.0%). Chemotaxonomic analyses indicated that the main respiratory quinone in both strains is ubiquinone-10. The DNA G+C contents of strains TB0136T and TB0139T were 63.3 and 61.5 mol%, respectively. Phylogenetic analysis based on the 16S rRNA gene sequence and the whole genome indicated that strains TB0136T and TB0139T formed a unique lineage in the evolutionary branches of the family Xanthobacteraceae. According to the characteristics of polyphasic taxonomy, strains TB0136T and TB0139T are proposed to represent two novel species of a new genus, for which the name Haloleucobacter gen. nov. is proposed. The proposed species names are Haloleucobacter ellipsoideus and Haloleucobacter albus. The type strains are TB0136T (=MCCC 1K09187T=JCM 37006T) and TB0139T (= MCCC1K09185T=JCM 37007T), respectively.
