TL;DR

It is demonstrated that LYC protects mammary epithelial cells from FFA-induced oxidative damage via regulating TFEB-mediated autophagy, suggesting its potential use in alleviating metabolic stress in periparturient dairy cows.

Abstract

High concentrations of free fatty acids (FFA) during early lactation cause metabolic stress and oxidative damage in bovine mammary epithelial cells. This study explored the protective mechanism of lycopene (LYC), a natural antioxidant, against FFA-induced injury in the MAC-T cells. We found that 1.2 mM FFA induced oxidative stress, reactive oxygen species (ROS) accumulation, apoptosis, and impaired autophagy. LYC dose-dependently alleviated FFA-induced oxidative damage, restored autophagy flux, upregulated ATG5 autophagy-related 5 (ATG5), LC3-II, and transcription factor EB (TFEB), and downregulated p62, ROS, and apoptosis. Inhibition of autophagy abolished the protective effects of the LYC. Overexpression of TFEB mitigated FFA-induced dysfunction, while TFEB knockdown aggravated cell injury. These results demonstrate that LYC protects mammary epithelial cells from FFA-induced oxidative damage via regulating TFEB-mediated autophagy, suggesting its potential use in alleviating metabolic stress in periparturient dairy cows.

Role of endoplasmic reticulum stress-mediated autophagy in cadmium-induced apoptosis in BRL-3A cells

Cadmium (Cd), a widely present environmental toxicant, can cause liver damage following chronic exposure. Although endoplasmic reticulum stress (ERS) and autophagy are known to be involved in Cd-induced liver injury, their molecular mechanisms remain incompletely understood. In this study, using an in vitro Cd exposure...

The crosstalk between sequestosome 1 and heme oxygenase-1 in regulating autophagy/oxidative stress in bovine mammary epithelial cells.

Bovine mastitis severely compromises dairy cattle health, primarily through oxidative stress and dysregulated autophagy in bovine mammary epithelial cells (BMECs), ultimately impairing lactation performance. While heme oxygenase-1 (HO-1) is well-established as a critical regulator of oxidative stress and autophagy, its...

SALVIANOLIC ACID B ATTENUATES GLUCOLIPOTOXICITY-INDUCED OXIDATIVE STRESS AND MITOCHONDRIAL DYSFUNCTION IN AML12 HEPATOCYTES VIA MODULATING CRBN/AMPK SIGNALING

It is suggested that SalB protects hepatocytes from glucolipotoxic injury by limiting oxidative damage, preserving mitochondrial function, and restoring CRBN-associated AMPK signaling.

S‐Equol Alleviates Palmitic Acid–Induced Muscle Atrophy by Modulating the ERβ–AMPK–Autophagy Axis in L6 Cells

Background Diabetic sarcopenia arises from metabolic imbalance–induced ROS accumulation and oxidative stress, which exacerbate muscle degeneration. Typically, autophagy removes excessive ROS and damaged organelles, helping to limit oxidative stress and preserve skeletal muscle function. S‐Equol (Eq), an intestinal meta...

ROS-mediated autophagy-apoptosis crosstalk regulates cell fate in the marine microalga Isochrysis galbana under BDE-47 stress.

Although the regulatory crosstalk between autophagy and apoptosis in response to stress has been well documented in higher multicellular eukaryotes, its functional significance in marine microalgae exposed to persistent organic pollutants (POPs) remains largely unclear. Here, we investigated the coordinated responses o...

AgeViva Modulates Inflammatory Responses, Autophagy, and Mitochondrial Homeostasis in BV-2 Cells and C. elegans

Background: Oxidative stress, neuroinflammation, impaired autophagy, and mitochondrial dysfunction are major contributors to ageing and neurodegenerative diseases. This study investigated whether AgeViva could counteract these processes by modulating redox balance, inflammation, autophagy, and mitochondrial function. M...
