The findings support the potential application of phage therapy as an alternative therapeutic treatment against lung infections caused by carbapenem-resistant A. baumannii and suggest that they are potential candidates for phage therapy.

Abstract

Abstract Acinetobacter baumannii has raised significant concern due to its high rates of morbidity and mortality, coupled with its increasing resistance to multiple antibiotics including carbapenems. In this current study, two new lytic phages Srynta and Nolivar were isolated from sewage water, characterized in vitro and their therapeutic potential was assessed using a mouse model of pneumonia. In vitro results showed that both phages show efficient adsorption to the propagating host, have good lytic activity, short latent period and high burst size. Sequence analysis showed that Srynta and Nolivar phages have linear dsDNA genomes of 45,218 and 41,230 base pairs with G+C content of 38 mol% and 37.9 mol% and contain 88 and 83 coding sequences, respectively. Transmission electron microscopy showed the icosahedral capsid and contractile tail of the phages and genomic analysis predicted that both phages have myovirus morphology and belong to the class Caudoviricetes. Genomic analysis suggested the therapeutic safety of these phages, as they do not have bacterial virulence factors, antimicrobial resistance genes or lysogenic genes, suggesting that they are potential candidates for phage therapy. The in vivo study demonstrated a high survival rate in the phage cocktail treatment group compared to the control group. Our findings support the potential application of phage therapy as an alternative therapeutic treatment against lung infections caused by carbapenem-resistant A. baumannii.

Preliminary in vitro characterization and lytic dynamics of three novel siphovirus-like bacteriophages targeting extensively drug-resistant Vibrio alginolyticus V79

The findings suggest that, while all three phages exhibit high environmental resilience, Phage P25 is the most promising candidate for further genomic safety validation, serving as a vital biological component in therapeutic phage cocktail formulations to manage XDR vibriosis in sustainable aquaculture.

Isolation and Characterization of Lytic Bacteriophages Targeting Clinical Staphylococcus Other Than S. aureus.

Staphylococcus spp., other than S. aureus (SOSA, formerly CoNS), are opportunistic pathogens often linked to biofilm infections on indwelling devices. Their rising antibiotic resistance highlights the need for novel therapies. Bacteriophages are promising due to their specificity and ability to inhibit cell growth and...

Genomic Characterization of 30 Lytic Klebsiella pneumoniae Bacteriophages

The emergence and spread of Gram-negative pathogens that are resistant to multiple antibiotic classes pose a global risk to public health. Among such pathogens, carbapenem-resistant strains of Klebsiella pneumoniae (CR-KP) are of concern due to their resistance to treatment by most antibiotics and the mobility of their...

Characterization of Escherichia Phage ND-4 Against Avian Pathogenic Escherichia coli IMT5155 and Its Preliminary Application in Chicks

Colibacillosis caused by avian pathogenic Escherichia coli (APEC) remains an important bacterial disease in poultry production and is increasingly difficult to control because of antimicrobial resistance. In this study, a lytic phage, ND-4, was isolated from poultry-farm sewage using the O2:K1 APEC strain IMT5155 as th...
