It is demonstrated that salt and osmotic stresses rapidly increase cytosolic Ca2+ in imbibed seeds, and that the calcium sensor CALMODULIN-LIKE 20 (CML20) is crucial for germination tolerance to these stresses.

Abstract

Plants integrate multiple environmental cues to decide the timing of seed germination. Light promotes germination primarily through photoreceptor phytochrome B (phyB), which eliminates the action of transcription factor PHYTOCHROME-INTERACTING FACTOR 1 (PIF1). Conversely, abiotic stresses typically elevate cytosolic Ca2+ and inhibit germination. However, the coordination of light and calcium signaling during germination under stress conditions remains poorly understood. Here, we demonstrate that salt and osmotic stresses rapidly increase cytosolic Ca2+ in imbibed seeds, and that the calcium sensor CALMODULIN-LIKE 20 (CML20) is crucial for germination tolerance to these stresses. CML20 directly interacts with PIF1 in a Ca2+-dependent manner, suppressing PIF1 self-association, oligomerization, and DNA-binding capacity. Moreover, phyB interacts with CML20 and PIF1 independently, promoting the assembly of the CML20-PIF1 complex and further reducing PIF1’s transcriptional activity. Genetic analyses reveal that PIF1 functions downstream of CML20, and that CML20 and phyB operate within the same pathway. Together, our findings define a phyB-CML20-PIF1 module, providing a rapid, reversible, and non-proteolytic mechanism to optimize seed germination under stress conditions. Plants integrate multiple environmental cues to time seed germination. Here, the authors show that the calcium-sensing CML20 protein links light and stress signals, enabling seeds to germinate under salt and drought conditions.

The BEE2 gene integrates brassinosteroid signalling into a regulatory module controlling gibberellin-mediated germination

Results indicate that BEE2 seems to provide an additive function to the RGL2-NAC-EXPA2 regulatory module to control the germination process mainly when GAs levels are compromised, which adds an extra layer of complexity to the control of the germination process.

A NAC-MYB-cysteine regulatory module enhances drought and salt tolerance in soybean.

Drought and salinity are two major factors limiting soybean productivity worldwide, and both stresses can negatively affect plant growth in similar manners by disrupting cellular redox homeostasis and metabolism. However, the regulatory networks coordinating transcriptional control and metabolic adaptation under both t...

Abscisic acid–regulated stability of CmABF1 and CmBRM modulates salt tolerance in chrysanthemum via epigenetic regulation of CmHSFA4

It is shown that salt stress–induced ABA accumulation up-regulates Heat Shock Factor 4 (CmHSFA4), a gene that is known to enhance chrysanthemum salt tolerance, and an ABA-SnRK2.2-ABF1/BRM signaling module that integrates phosphorylation-dependent protein stabilization and degradation with histone methylation dynamics t...

Cytokinin Mediates Photosynthesis and Senescence Action Through CHLOROPLAST IMPORT APPARATUS 2 (CIA2) and CIA2-like (CIL)

Progression of leaf senescence is regulated by internal factors like the plant hormone cytokinin (CK) that acts to delay senescence and retain photosynthesis by maintaining chloroplast-functionality. While initial CK-signaling components controlling senescence have been described, downstream targets regulating physiolo...
