TL;DR

It is shown that temporally resolved transcriptional regulatory networks integrating hormonal crosstalk underlie cold-induced modulation of seed dormancy and germination in wheat and that jasmonate (JA)-gibberellin (GA) synergism and JA-abscisic acid (ABA) or GA-ABA antagonism play roles in regulating dormancy release during imbibition of cold-treated dormant seeds at OT.

Abstract

Using a combination of genome-wide transcriptome, targeted hormonome and pharmacological approaches, this study showed that temporally resolved transcriptional regulatory networks integrating hormonal crosstalk underlie cold-induced modulation of seed dormancy and germination in wheat. Gene homoeologues (GHs) differentially expressed between cold-treated and untreated dormant seeds imbibed at optimal temperature (OT, 22°C) as well as between the same seed samples imbibed in cold and at OT represent distinct biological processes (BPs), indicating the role of coordinated activation/repression specific BPs in cold-mediated alteration of dormancy and germination. Our analyses highlighted that jasmonate (JA)-gibberellin (GA) synergism and JA-abscisic acid (ABA) or GA-ABA antagonism play roles in regulating dormancy release during imbibition of cold-treated dormant seeds at OT. The repression of germination in dormant seeds imbibed in cold compared to those imbibed at OT is associated with synergistic repression of seed sensitivity to JA and GA and activation of its sensitivity to ABA. Specific members of TaWRKY, TabHLH, TaMYB, TaNAC, TaERF, TaARF and TabZIP transcription factor families establish the transcriptional network involving specific GA, ABA and JA metabolism and signaling GHs and thereby the ABA-GA-JA crosstalk that enhance dormancy release during imbibition at OT after cold treatment and inhibition of germination during cold treatment.

Response of Dormancy-associated MADS-box (DAM) and Flowering Locus T (FT) genes to contrasting temperature conditions during dormancy in apricot (Prunus armeniaca L.).

Chilling requirement (CR) fulfilment is critical for successful flowering in temperate fruit trees. In mild-winter regions, several anomalies can occur when the cultivar-specific CR is not fully met, including irregular budbreak, bud abortion, and ultimately reduced fruit set. In this study, we examined the molecular r...

Multi-hormonal crosstalk regulating seed dormancy, germination and plant growth: Integration of abscisic acid, gibberellins, jasmonic acid and salicylic acid signalling

Phytohormones are small molecules that act as signalling molecules in plant growth and development, seed dormancy, seed germination and environmental stress responses and they interact with one another through interconnected signalling pathways. Among these, abscisic acid (ABA) and gibberellins (GA) have antagonistic f...

Negative regulation of seed germination in Medicago truncatula by the lncRNA MtCIR1 under salt stress is associated with alternative splicing.

Seed germination, an initial stage of the plant life cycle, is sensitive to salt stress. Phytohormones such as abscisic acid (ABA) and gibberellins (GA) are involved in the suppression of seed germination by salt stress. Despite identification of numerous salt stress-responsive long noncoding RNAs (lncRNAs) in differen...

Germination-Related Universal Stress Protein (GRUSP) Is Involved in Abscisic Acid Responses During Imbibition and Early Post-Germination Growth of Arabidopsis Seeds

The universal stress protein (USP) family is a large but poorly studied group of plant proteins whose roles are considered primarily in the context of stress tolerance. This study demonstrates that the functions of USPs are not limited to the stress response but also influence plant developmental processes, namely, the...

The miR1866-OsUBP7 regulatory module promotes grain yield and seed dormancy through abscisic acid signaling in rice.

Higher grain weight and stronger seed dormancy are key objectives for improving rice (Oryza sativa) yield and inhibiting pre-harvest sprouting. Therefore, identifying genes that coordinately regulate grain weight and seed dormancy is an urgent priority. Here, we report that knocking out miR1866 (KO1866) increased grain...
