A comprehensive review of immunotherapy for NPC is provided, evaluating current progress an persistent challenges by drawing insights from both completed and ongoing clinical trials.

Abstract

ABSTRACT Immunotherapy has emerged as a cornerstone in the management of both recurrent/metastatic nasopharyngeal carcinoma (R/M NPC) and locoregionally advanced NPC (LANPC). Evidence indicates that programmed death-1/programmed death ligand-1 (PD-1/PD-L1) inhibitors, whether administered as monotherapy or in combination with chemotherapy or antiangiogenic agents, can significantly improve outcomes in R/M NPC. For LANPC, incorporating immunotherapy into induction, concurrent chemoradiotherapy, and adjuvant phases has demonstrated potential benefits in enhancing antitumor immune responses and reducing the incidence of disease recurrence. However, challenges persist regarding the identification of optimal patient populations, the determination of optimal timing and duration of treatment, and the management of immune-related adverse events. Biomarkers such as PD-L1 expression and plasma Epstein-Barr virus-DNA levels show promise in facilitating risk stratification and tailoring individualized treatment regimens. Accordingly, this review provides a comprehensive review of immunotherapy for NPC, evaluating current progress an persistent challenges by drawing insights from both completed and ongoing clinical trials.

Systemic therapy for metastatic nasopharyngeal carcinoma in the immunotherapy era: A critical review.

BACKGROUND The prognosis of metastatic nasopharyngeal carcinoma (mNPC) remains dismal despite recent advances of locoregional and systemic therapy. The incorporation of programmed death-1 (PD-1) inhibitors into first-line chemotherapy has transformed the therapeutic landscape; however, optimal treatment strategies beyo...

Current Perspectives on the Use of Adjuvant Pembrolizumab for Hepatocellular Carcinoma

Abstract Despite implementation of surveillance in at-risk populations and the availability of potentially curative treatment such as resection or ablation, recurrence of hepatocellular carcinoma (HCC) remains a frequent clinical challenge driven by both occult residual disease and de novo carcinogenesis, and can have...

Guiding the Application of Immunotherapy in Nonsmall Cell Lung Cancer: The Role of Biomarkers

Although immunotherapy has completely transformed the treatment landscape of nonsmall cell lung cancer (NSCLC), its wide application is still limited by the heterogeneity of patient responses, primary and acquired resistance, as well as the management challenges of immune‐related adverse events (irAEs). To achieve prec...
