TL;DR

The dual-target c-Met/VEGFR kinase inhibitor foretinib reshapes tumor immune microenvironment (TIME) by inducing immunogenic cell death (ICD) and the combination of foretinib with anti-CD47 antibody significantly inhibits tumor growth in colon cancer model.

Abstract

Highlights • The dual-target c-Met/VEGFR kinase inhibitor foretinib reshapes tumor immune microenvironment (TIME) by inducing immunogenic cell death (ICD).• We identified an on-target mechanism of foretinib in transcriptionally activating Drath Receptor 5 (DR5), a key component of the extrinsic apoptotic pathway.• The combination of foretinib with anti-CD47 antibody significantly inhibits tumor growth in colon cancer model.

TREM1 Blockade Reprograms Tumor‐Associated Macrophages and Improves Anti‐PD‐1 Efficacy in Gastric Cancer

The objective response rate of immune checkpoint inhibitors in patients with gastric cancer is limited. Targeting specific subtypes of macrophages holds promise for expanding the potential population sensitive to immune checkpoint inhibitors. This study aims to evaluate the key role of TREM1+tumor‐associated macrophage...

Antineoplastic Drugs: Research Progress of Immune Checkpoint CD47 Inhibitors.

Immunotherapy using immune checkpoint inhibitors represents a breakthrough in cancer treatment. The overexpressed CD47 on tumor surfaces functions as a 'don't eat me' signal by binding to signal regulatory protein alpha (SIRPα) on macrophages, facilitating immune escape. However, the advancement of targeted CD47 inhibi...

STAT3 inhibition for the induction of immunogenic cell death in multiple myeloma

It is reported that STAT3 inhibition eliminates multiple myeloma stem-like cells while eliciting hallmarks of immunogenic cell death, yet in vivo validation remains necessary before this strategy can be considered immunogenic.

TNFR1 expression on cancer cells mediates immune escape and an immunosuppressive microenvironment

Evidence is provided for a key role of TNFR1, as expressed by malignant cells, in orchestrating a pro-tumor immunosuppressive environment through secondary inflammatory mediators through secondary inflammatory mediators.
