TL;DR

It is demonstrated that thermoneutral housing markedly accelerates Western diet-induced metabolic dysfunction-associated steatohepatitis (MASH), resulting in enhanced hepatic inflammation, fibrosis, and disease severity, and it is established that providing homeostatic conditions via thermoneutral housing is an important element and a practical strategy for improving the translational relevance of preclinical MASH models.

Abstract

In this issue, Leke et al. demonstrate that thermoneutral housing markedly accelerates Western diet-induced metabolic dysfunction-associated steatohepatitis (MASH), resulting in enhanced hepatic inflammation, fibrosis, and disease severity. Importantly, thermoneutral conditions produce a hepatic transcriptional landscape that more closely resembles human MASH than conventional housing. These findings further establish that providing homeostatic conditions via thermoneutral housing is an important element and a practical strategy for improving the translational relevance of preclinical MASH models.

Phosphodiesterase 4 Inhibition Attenuates Diet-Induced Obesity and Activates BAT Thermogenic Pathways Through Central Mechanisms.

ARTICLE HIGHLIGHTS Metabolic alterations in obesity encompass impaired thermogenesis and sympathetic activity, underscoring the need for new metabolic targets to treat this disease. This study tested whether inhibition of PDE4 by rolipram enhances energy expenditure and brown adipose tissue activity. Rolipram reduced b...

An RNA thermogenic therapy to preserve lean mass and enhance metabolic health during GLP-1 weight loss

Significance Current antiobesity therapies reduce body weight but often induce loss of lean mass and fail to achieve durable metabolic improvements after treatment ends. We show that an adipose-targeted RNA therapeutic targeting ZFP423, a transcriptional repressor of brown and beige fat identity, enhances thermogenesis...

Early-life stress-induced gut dysbiosis is ameliorated by nicotinamide treatment

A nicotinamide-driven amelioration of gut dysbiosis in middle-aged rodents with a history of neonatal maternal separation is reported, demonstrating nicotinamide-driven amelioration of gut dysbiosis in middle-aged rodents.

Repeated Moderate and Severe Heat Exposure Has Divergent Effects on Skeletal Muscle Mitochondrial, Proteostatic and Neuromuscular Outcomes in Mice

This work examined whether repeated moderate (39°C) and severe whole‐body hyperthermia produce distinct skeletal muscle trajectories involving mitochondrial dynamics, proteostasis, contractile gene programming, and motor behavior.

Einfluss des Darmmikrobioms auf Gesundheit und Alterung

During aging, the gut microbiome undergoes profound compositional and functional changes that may compromise host health. Using metabolic modeling and mouse studies, we showed that aging is associated with reduced microbial cooperation, lower production of beneficial metabolites, accumulation of pro-inflammatory comp...

Beyond BMI: The diet-microbiome context of the obesity paradox.

In Nature, Desharnais et al. show that in diverse mouse models, anti-PD-1 sensitivity is more closely linked to the diet-microbiome-metabolite ecosystem than to obesity-associated metabolic dysfunction. Their findings reinforce body mass index (BMI) as an incomplete proxy for host biology and suggest strategies to impr...
