Abstract

Marine invertebrates are an important reservoir of microbial diversity, yet the culturable bacteria associated with them remain poorly studied. This study describes the isolation and comprehensive characterization of bacterial isolates associated with invertebrates of the South China Sea. Of the 40 isolates obtained, five strains differing in morphological characteristics were selected for further study; their biochemical properties and cell ultrastructure were examined, and phylogenetic analysis was performed. Phylogenetic analysis based on partial 16S rRNA gene sequences revealed that the strains belong to the classes Alphaproteobacteria and Gammaproteobacteria, with the greatest similarity to representatives of the genera Roseibium, Halomonas, Alteromonas, and Pseudoalteromonas. Biochemical characterization (API 20E and ID32 test systems) revealed a limited ability of the isolates to utilize a number of substrates, alongside lipase and β-glucosidase activity. Ultrastructural studies revealed a cell wall structure characteristic of gram-negative bacteria, as well as vesicle-like structures in TEM images. Several isolates had a surface polysaccharide layer, and one strain (5-1g) had polysaccharide filaments presumably associated with the formation of cellular conglomerates. The results obtained expand the understanding of the morphological, biochemical, and phylogenetic diversity of cultured bacteria associated with marine invertebrates and form the basis for further, targeted study of their role in host association.
