Genetic fidelity of micropropagated shoots from recalcitrant seeds of the medicinally important tree: Garcinia mangostana
Abstract
Garcinia mangostana Linn is a medicinally important and commercial fruit-bearing tropical tree. The traditional horticultural methods for propagation have had little success in meeting the growing demand for the fruit. Micropropagation is a method of plant tissue culture for the mass production of plants. The surfacesterilized seeds were a source of axenic explants. The Murashige Skoog base medium was supplemented with several Plant Growth Regulators (PGR). The best seed germination response was seen with MS half-strength basal media supplemented with 4 µM BAP. The combination of MS half-strength + 4 µM BAP + 5 µM IAA produced the highest shoot number and broke bud dormancy the fastest. The best rooting response was provided by MS half-strength + 3 µM NAA. The outcomes showed that these techniques could be applied for multiple shoot induction from the seed to produce the plant in large quantities. The in vitrogrown explants were 97.78% true to type after testing for somaclonal variations using ISSR primers (AG)8T, (AG)8AA and (GA)9C.
