Evidence that virus-derived genomic elements may acquire developmental functions after endogenisation is provided, expanding the understanding of the functional domestication of endogenous viral elements and their potential contribution to the developmental and ecological adaptation of rice planthoppers.

Abstract

Non-retroviral endogenous viral elements (nrEVEs) are viral-derived sequences integrated into host genomes, but the biological functions of Chuvirus-derived nrEVEs (ChuEVEs) in insects remain poorly understood. Here, we identified homologous ChuEVEs in the genomes of rice insect vectors Sogatella furcifera and Laodelphax striatellus, designated SfChuEVE and LsChuEVE. SfChuEVE and LsChuEVE showed similarity to the glycoprotein of Sanya chuvirus 2 and were detectable in most examined individuals. Transcript analysis revealed that ChuEVEs were transcriptionally active and contained predicted ORFs with distinct coding organisations. Both ChuEVEs were transcriptionally active across developmental stages and were highly expressed in salivary glands. RNAi-mediated knockdown of SfChuEVE or LsChuEVE significantly prolonged nymphal development and altered genes involved in the 20-hydroxyecdysone (20E) and insulin/insulin-like signalling/target of rapamycin (IIS/TOR) pathways in both planthopper species. Further silencing of IIS/TOR components, including S6K and FOXO, affected selected 20E pathway-related genes, particularly USP and EcR, whereas silencing IIS/TOR components did not alter ChuEVE transcription. Moreover, exogenous 20E restored the expression of representative 20E signalling genes but failed to rescue the reduced expression of IIS/TOR-related genes following ChuEVE knockdown. Together, these findings support a role for ChuEVEs in nymphal development through an IIS/TOR-20E regulatory axis. Our study provides evidence that virus-derived genomic elements may acquire developmental functions after endogenisation, expanding our understanding of the functional domestication of endogenous viral elements and their potential contribution to the developmental and ecological adaptation of rice planthoppers.

An RNAi-Based Vulnerability in the White-Backed Planthopper: Silencing SfHSP70-8 Impairs Thermotolerance and Insecticide Susceptibility in a Key Rice Pest

Heat shock protein 70 (HSP70) serves as a critical molecular chaperone governing environmental stress responses in insects. This study aimed to characterize the expression pattern of SfHSP70-8 and determine its functional role in thermotolerance and insecticide susceptibility in the white-backed planthopper, Sogatella...

Botrytis virus X ORF2 suppresses RNA silencing in a trigger-restricted manner and is associated with altered Dicer-like gene expression in Botrytis cinerea

This study identifies Botrytis virus X (BVX) ORF2 (X2) as a trigger-restricted suppressor that enhances GFP accumulation in plant-based assays initiated by sense RNA but does not suppress silencing initiated by defined hairpin-derived siRNA or miRNA-guided pathways.

Lmx1b is essential for molting and leg integrity in Henosepilachna vigintioctopunctata: Implications for RNA biopesticide development.

LIM homeodomain transcription factor 1 beta (Lmx1b), a member of the LIM-homeodomain family, plays essential roles in tissue differentiation and organ development in diverse eukaryotes, yet its biological functions in insects remain largely unknown. Here, we identify Lmx1b as a highly effective RNA interference (RNAi)...

Five novel ubiquitous totiviruses function as virulence-promoting symbionts in the obligate biotrophic fungus Puccinia triticina

It is demonstrated that totiviruses function as virulence-promoting symbionts in Pt, establishing for the first time a functional link between totiviral infection and enhanced pathogenicity in cereal rust fungi and identifying candidate targets for RNAi-based disease control.
