Formulation and Evaluation of Nanoparticle-Loaded Mucoadhesive Buccal Film of Ondansetron
Abstract
This study aimed to formulate and evaluate an ondansetron hydrochloride-loaded nanoparticle mucoadhesive buccal film to enhance buccal drug delivery and potentially reduce the limitations associated with conventional oral administration. Ondansetron hydrochloride was incorporated into chitosan nanoparticles using the ionic gelation method with sodium tripolyphosphate as the cross-linking agent. The prepared nanoparticles were incorporated into a mucoadhesive film composed of hydroxypropyl methylcellulose and polycarbophil using the solvent casting method, with glycerol as a plasticizer. The prepared nanoparticle-loaded buccal films were evaluated for appearance, weight variation, thickness, surface pH, folding endurance, mechanical properties, swelling index, mucoadhesive strength, drug content uniformity, surface morphology, in-vitro drug release, and ex-vivo permeation. The developed formulation was intended to provide a uniform, flexible, and mucoadhesive buccal delivery system for ondansetron hydrochloride. The nanoparticle-loaded buccal film showed potential for prolonged residence at the buccal mucosa and improved drug delivery. Further in-vivo studies are required to establish its effect on bioavailability and therapeutic performance.
