TL;DR

It is demonstrated that PEZO-1 structural integrity is essential for vesicle formation and transport; truncating either the N-terminal transmembrane domains or the C-terminal ion pore induces aberrant vesicle morphology and arrests trafficking.

Visualizing Protein Trafficking in Whole Tissue Context Using Super-Resolution Nanoscopy in Drosophila.

The fruit fly Drosophila melanogaster is widely used to study conserved developmental and cellular signaling pathways in vivo. Many signaling pathways, including Wnt signaling, depend on tightly regulated secretion, endocytosis, and intracellular trafficking events. However, visualizing the dynamics of membrane-associa...

Sperm development requires TMC5-dependent mechanoresponsive calcium entry

Spermiogenesis, the transformation of round spermatids into streamlined, fertilization-competent sperm, represents cellular morphogenesis at its most extreme1,2. Despite decades of effort, spermiogenesis remains difficult to recapitulate outside the testis2,3, highlighting the importance of cues within the native tissu...

Dynamic clustering and Rab11-dependent regulation of voltage-gated proton channels at the plasma membrane.

AIMS Voltage-gated proton channels (HV1) play a key role in intracellular pH homeostasis in immune cells and have been implicated in the neurological damage after a stroke and the prognosis and malignancy of several tumors. Thus, the molecular mechanisms governing HV1 surface abundance are of great clinical significanc...

The role of organelles as mechanotransducers.

Mechanobiology examines how physical forces influence cellular structure and function. Traditionally, mechanotransduction has been attributed to plasma membrane receptors, focal adhesions, and the cytoskeleton. However, emerging evidence highlights intracellular organelles as active contributors to mechanosensing and f...

Arrestins as programmable integrators of GPCR signaling: structural microstates, spatiotemporal logic, and therapeutic control

Recent structural, biochemical, and physiological insights are synthesized to outline how arrestins decode regulatory inputs into signaling outcomes and how this knowledge enables the development of next-generation, structure-guided GPCR therapeutics.

Cargo crosslinking nucleates AP-1 lattice formation.

Cryo-electron microscopy structures of the human AP-1 complex bound to a cargo containing tandem sorting motifs reveal that a single cargo molecule bearing tandem motifs can bridge two neighboring AP-1 complexes through a previously unrecognized μ1-σ1 inter-complex interface, thereby promoting adaptor oligomerization.
