Formamidine Chemistry: Recent Synthetic Developments and Functionalization Strategies
Abstract
Formamidines represent a very important class of nitrogen‐containing organic substances with various applications in synthetic organic chemistry, as well as in medicinal chemistry, pharmaceuticals, agrochemistry, and functional materials. The article summarizes scientific advances in the use of formamidine frameworks for synthesis and applications from 2016 to 2025. Acid‐catalyzed condensation reactions, multicomponent approaches, metal‐free methods, and transition‐metal‐catalyzed methods are major techniques used for the synthesis of formamidines. Special emphasis is placed on topical modifications of formamidines, which enhance their diversity and make them more applicable to biological and materials science purposes. The article provides an overview of the methods used for the development of synthetic formamidines and analyzes their practical applications in synthesis. Various mechanistic studiesin the field address the peculiarities of formamidine synthesis, while substrate scope, selectivity, and reaction conditions must be taken into account for scientific work. Examples of N ‐heterocycles demonstrate the efficiency of the compounds discussed in medicine, leading to the successful use of formamidines in drug development and pesticide production.
