A hierarchical transcription-polyadenylation axis that associate with placental trophoblast syncytialization is revealed and fill key gaps in mRNA processing and provide a foundation for studying the transcriptional regulatory mechanisms underlying trophoblast differentiation.

Abstract

Polyadenylation, a core transcriptional step governed by polyadenylation signals (PAS), functionally interacts with transcription. Nevertheless, how PAS sequences respond to variable transcriptional output remains poorly characterized. Here, we precisely modulated cellular transcription levels by integrating SV40 promoter truncation with a Tet-On dual-luciferase reporter system. Diminished polyadenylation efficiency accompanies lowered promoter activity, and graded transcription assays revealed a tiered PAS processing model: canonical and suboptimal PASs maintain stable 3′-end processing at high transcription levels, whereas non-canonical PASs display compromised processing efficiency with rising transcription. During forskolin(FSK)-induced BeWo cell syncytialization, endogenous trophoblast fusion genes carrying potent canonical PASs were selectively mobilized upon transcriptional activation to enrich differentiation transcripts and sustain orderly syncytiotrophoblast maturation. Together, these findings reveal a hierarchical transcription-polyadenylation axis that associate with placental trophoblast syncytialization. They fill key gaps in mRNA processing and provide a foundation for studying the transcriptional regulatory mechanisms underlying trophoblast differentiation.

Temporal dynamics of intragenic enhancer RNAs regulate Lin28a expression in response to external stimuli in embryonic stem cells

Abstract LIN28A is a conserved RNA-binding protein essential for pluripotency, let-7 miRNA regulation, and cellular metabolism. However, the mechanisms controlling its transcriptional activation during early stem cell fate transitions remain poorly defined. Using the Lin28a locus as a model, we identified multiple intr...

Cis-element acquisition converts pervasive transcripts into translated mRNAs

The mammalian genome is pervasively transcribed into thousands of unstable non-coding RNAs, including promoter-upstream transcripts (PROMPTs) and enhancer RNAs (eRNAs). These pervasive transcripts are degraded by the nuclear exosome within minutes of synthesis and are never translated. Coding and non-coding transcripts...

Proximity of pre-mRNA 3' end processing and transcription termination is associated with enhanced gene expression.

Two key factors required for pre-mRNA 3' end processing, CPSF73 and PCF11, are linked to poor cancer prognosis, yet their oncogenic roles remain unclear due to their dual function in transcription termination. Using a colorectal cancer (CRC) progression model, we show that primary tumor cells depend on high CPSF73 and...

Post-transcriptional regulation of Profilin-2 by microRNAs and RNA-binding proteins forms a critical regulatory node for early embryonic cell fate decisions

It is demonstrated that post-transcriptional regulation of the Profilin 2 (Pfn2) transcript is essential for differentiation of embryonic stem cells into the primary germ layer lineages and revealed the Pfn2 microRNA-IRE regulatory axis as a critical post-transcriptional regulatory node governing the switch from plurip...

Dynamic Polycomb-CBC crosstalk orchestrates mRNA production at transcriptionally active loci in plants.

It is shown that in Arabidopsis thaliana, the chromodomain H3K27me3 reader LIKE HETEROCHROMATIN PROTEIN1 binds nascent precursor mRNA through its middle intrinsically disordered region at H3K27me3-coated chromatin loci, thereby attenuating transcription efficiency.
