It is demonstrated that USP26 functions as an oncogenic driver in NSCLC by stabilizing Gαs and suppressing autophagy, highlighting its potential as a prognostic biomarker and therapeutic target.

Abstract

Non-small cell lung cancer (NSCLC) remains the leading cause of cancer-related mortality worldwide. Ubiquitin-specific protease 26 (USP26) has been implicated in tumor progression, but its functional role and molecular mechanism in NSCLC remain poorly understood. USP26 was significantly upregulated in NSCLC tissues and cell lines, correlating with poor prognosis. Functionally, USP26 promoted cell proliferation and colony formation while inhibiting apoptosis. Mechanistically, USP26 directly interacted with and deubiquitinated Gαs, stabilizing it to activate the cAMP/PKA signaling pathway. USP26 also suppressed autophagic flux and ER-phagy, and its pro-tumor effects were abrogated by the autophagy inhibitor 3-MA. Our findings demonstrate that USP26 functions as an oncogenic driver in NSCLC by stabilizing Gαs and suppressing autophagy, highlighting its potential as a prognostic biomarker and therapeutic target.

The deubiquitinating enzyme USP42 promotes lung adenocarcinoma progression through PTGS2

The ubiquitin-specific peptidase 42 (USP42) is a cysteine-dependent deubiquitinase with pro-tumorigenic roles in several cancers. In lung adenocarcinoma (LUAD), elevated USP42 expression correlates with advanced stage and poor survival, mirroring the prognostic impact of prostaglandin-endoperoxide synthase 2 (PTGS2) ov...

SET Oncoprotein in Gastric Cancer: A Master Regulator of PP2A Suppression and Tumor Progression

Therapeutic modulation of the SET-PP2A axis may provide a novel strategy for suppressing tumor progression and improving treatment outcomes, and further mechanistic studies and welldesigned clinical investigations are required to establish the therapeutic relevance of SET targeting and facilitate its integration into p...

GPX3 suppresses lung adenocarcinoma progression through HSPB1 ubiquitination and Hippo pathway activation.

A robust prognostic model based on manganese metabolism-related genes is established and the critical regulatory role of the GPX3/HSPB1/yes-associated protein (YAP) axis in LUAD progression is uncovered, underscoring the therapeutic potential of targeting GPX3.

TRIM46 drives lung adenocarcinoma progression by stabilizing PRDX1 via SUMOylation

Lung adenocarcinoma (LUAD) is one of the major subtypes of non-small cell lung cancer (NSCLC), but the molecular mechanisms underlying LUAD initiation and progression remain incompletely understood. In the present study, we identified TRIM46 as a key driver of LUAD progression. TRIM46 is significantly overexpressed i...
