Environmental DNA (eDNA) metabarcoding can provide fish-community inventories across river networks. We surveyed ten sites in the Jingdezhen section of the Raohe River system on 20–23 October 2023 and 2–5 June 2024, covering the Donghe River, Chang River, and Le’an River. Three technical process replicates were pooled per site and campaign to produce 20 community profiles. The two surveys detected 96 and 94 taxa, respectively, including 100 fish taxa overall, of which 90 were shared. Richness, Shannon diversity, Simpson diversity, and Pielou evenness did not differ significantly between paired campaigns. Community composition differed between campaigns (Bray–Curtis PERMANOVA: R2 = 0.293; Jaccard: R2 = 0.185; both adjusted p = 0.002). PERMDISP did not detect unequal dispersion. River-group differentiation was clearer in 2023, whereas neither distance metric was significant in 2024 after multiple-testing correction. A reduced environmental partial RDA accounting for site and campaign effects was not significant (F = 0.971, p = 0.420). These findings provide a two-campaign molecular inventory for the ten surveyed sites and identify between-campaign compositional differences, without demonstrating seasonal dynamics or independent environmental drivers.
Environmental DNA (eDNA) metabarcoding is an efficient, non-invasive approach for surveying fish diversity in complex habitats such as large rivers. This study conducted continuous eDNA monitoring along the Jiangsu section of the Yangtze River mainstem from 2022 to 2024 to characterize spatiotemporal variations in fish...

Reliable fish biodiversity baselines are essential for monitoring and adaptive management in large subtropical reservoirs. Environmental DNA (eDNA) metabarcoding provides a non-invasive approach for assessing fish assemblages, but molecular baseline information remains limited for Zhelin Reservoir in the Poyang Lake...
It is demonstrated that eDNA metabarcoding enhanced species detectability, resolved temporal and spatial community distribution characteristics with higher environmental sensitivity, and effectively characterized complex littoral microhabitats by identifying specific indicator species.

Simple Summary Environmental DNA (eDNA) metabarcoding has emerged as a revolutionary tool for freshwater ecosystem conservation and biodiversity monitoring, yet its feasibility and applicability to benthic invertebrates still require further study. In this study, eDNA metabarcoding was employed to reveal the diversity...

This preliminary study explored the use of mitochondrial COI metabarcoding to compare broad taxonomic detection patterns obtained from pooled environmental DNA (eDNA) and tissue-derived zooplankton DNA (T-DNA) samples collected from three Malaysian coastal locations in Johor: Pulau Kukup, Pengerang, and Pulau Besar. Wa...

Hydropower development has emerged as a major driver of global environmental change, causing significant ecological damage to freshwater ecosystems and threatening aquatic biodiversity. However, the impact of cascade dams on fish communities in large river systems remains poorly understood, particularly in biodiversity...
