TL;DR

A transcriptome-wide transition underlying the decline in poststorage embryo viability with increasing egg storage durations is revealed, suggesting a shift from adaptive stress responses in embryos from 4-day-stored eggs to maladaptive stress responses in embryos from 14-day-stored eggs, characterised by oxidative stress, apoptosis, and severe cellular damage.

Abstract

After oviposition, hatching eggs are generally stored at the breeder farm and hatchery before incubation is initiated. In certain cases, egg storage duration is extended beyond 7 days, which is referred to as 'prolonged storage'. Prolonged storage is associated with lower hatchability, delayed hatching, and worse chicken quality at hatch, but the underlying embryonic cellular processes that occur during egg storage are not yet fully understood. In contrast to previous targeted studies on single pathways, the current study employs a whole-transcriptome analysis to investigate physiological responses of broiler embryos to egg storage duration. Gene expression levels from broiler embryos isolated from fresh eggs (on the day of oviposition; n = 10) and from eggs that had been stored at 18 °C for either 4 days (n = 5) or 14 days (n = 5) were compared. Data were analysed by using the CLC Genomics Workbench platform. Results showed that genes common between broiler embryos from 4-day- and 14-day-stored eggs were associated with the positive regulation of the p38 mitogen-activated protein kinase cascade, controlling the balance between cell survival and death under cellular stress. Broiler embryos from 4-day-stored eggs showed gene expression levels indicative of cell cycle arrest at the Gap 1 to Synthesis transition in the cell cycle and mild endoplasmic reticulum stress compared to broiler embryos from fresh eggs. Broiler embryos from 4-day-stored eggs showed gene expression levels indicative of active cell proliferation and repair of minor DNA lesions compared to broiler embryos from 14-day-stored eggs. Broiler embryos from 14-day-stored eggs also showed gene expression levels indicative of cell cycle arrest at the Gap 1 to Synthesis transition, and additionally gene expression levels indicative of oxidative stress, apoptosis, repair of severe DNA damage, and severe endoplasmic reticulum stress compared to broiler embryos from fresh eggs. Current findings suggest that egg storage duration induced a shift from adaptive stress responses in embryos from 4-day-stored eggs to maladaptive stress responses in embryos from 14-day-stored eggs, characterised by oxidative stress, apoptosis, and severe cellular damage. This study reveals a transcriptome-wide transition underlying the decline in poststorage embryo viability with increasing egg storage durations.

Large variability of preincubation storage temperature in chukar partridge eggs under normal storage duration: Impact on egg and hatching traits, and embryonic development

The effect of different storage temperatures on egg traits during storage and incubation, hatching characteristics and embryonic mortality in chukar partridge eggs stored under normal-term storage was investigated in this study. The study was designed as a 2 × 4 factorial arrangement with two storage durations (7 and 1...

Effects of incubation temperature and in ovo supplementation of glucose and vitamin D3 on eggshell temperature, embryonic mortality, hatchability, chick quality, and post-hatch performance in broiler chickens

Introduction Incubation temperature and embryonic nutrient availability are critical determinants of successful embryogenesis and early post-hatch performance in broilers. In ovo feeding has emerged as an effective strategy to enhance embryonic nutrient supply. However, the efficiency of nutrient utilization by develop...

Developmental and transcriptomic alterations in bovine blastocysts derived from oocytes of donors exposed to toxic tall fescue.

Fescue toxicosis, caused by the ingestion of ergot alkaloids present in endophyte-infected tall fescue (E + ), is associated with impaired reproductive performance in beef cattle. Because early embryonic development depends on maternally derived RNAs and proteins accumulated during oocyte maturation, disruption of oocy...

Patterns of egg production across the lunar cycle in broiler breeder hens under tropical conditions.

1. This study evaluated whether lunar periodicity was associated with variation in productive traits and mortality in commercial broiler breeder hens. Production records from Cobb 500 heavy breeder hens (n = 17,548), collected between 2016 and 2021 on a commercial farm, were analysed.2. The synodic lunar cycle was divi...

Impact of hatchery microclimate, breeder age and storage time on egg water loss in a tropical climate

This study aimed to evaluate the effects of incubator microclimate, broiler breeder age, and fertile egg storage duration on egg water loss in a commercial hatchery within a tropical climate. Data were collected from the environments (air-conditioned egg room and incubator) in order to perform microclimatic assessments...
