TL;DR

It is suggested that TerC is involved in the transport of Mn2+ and Ca2+ across the thylakoid membrane, and that the absence of this function contributes to the albinotic phenotype observed in the terc seedlings.

Abstract

The oxygen-evolving complex (OEC) of photosystem II (PSII) contains a Mn4CaO5 cluster that catalyzes the light-driven oxidation of water. In Arabidopsis thaliana, optimal PSII function requires the thylakoid membrane-localized manganese transporter PHOTOSYNTHESIS AFFECTED MUTANT 71 (PAM71). Here, we show that overexpression of TELLURITE RESISTANCE C (TerC) restores low chloroplast manganese levels, O2 evolution rates, and growth defects in PAM71 knock-out plants. A TerC knock-down allele (amiR-TerC) exhibits OEC-related phenotypic changes similar to those observed in pam71, including altered period-four S-state oscillations of the OEC and PSII charge-recombination energetics. Unlike pam71, amiR-TerC accumulates a photosystem I (PSI) subcomplex lacking the subunits PsaL, PsaH and PsaO, impairing the ability to perform state transition. Diminished PSII activity and alterations in PSI are more pronounced under low calcium conditions, and light-dependent Ca2+ accumulation in isolated thylakoids is reduced in amiR-TerC. When expressed heterologously, TerC suppresses the Mn2+- and Ca2+-sensitive phenotypes of yeast mutant strains Δpmr1 and Δpmr1Δgdt1, respectively. We suggest that TerC is involved in the transport of Mn2+ and Ca2+ across the thylakoid membrane, and that the absence of this function contributes to the albinotic phenotype observed in the terc seedlings.

Arabidopsis STN7/STN8 alleviates oxidative damages via different regulatory patterns under drought stress

SUMMARY STN7 and STN8 kinases phosphorylate several proteins of the photosynthetic apparatus in response to changes in environmental conditions. Phosphorylation is linked to the dynamic arrangement of both the 3D structure of the thylakoid membrane and thylakoid protein complexes, thereby controlling excitation energy...

Effects of high light and elevated temperature on the structure of photosystem II isolated from Arabidopsis thaliana

Photosystem II (PS II) is a large membrane-bound pigment-protein complex responsible for extracting electrons and protons from water during photosynthesis and releasing molecular oxygen into the atmosphere. While light is required to drive the photochemical reactions, high light and high temperature can be damaging to...

ACHT2 deactivates carbon assimilation during light-dark transition

In chloroplasts, photosynthetic efficiency relies on a delicate balance between reductive and oxidative thiol-based signaling networks. Members of the high-midpoint-potential atypical thioredoxins (Trxs) were shown to oxidize photosynthetic enzymes by channeling reducing equivalents to H2O2 through 2-Cys-Prx activity....

β-Carbonic anhydrases integrate CO2 and light cues to coordinate photosynthesis in tomato.

Optimization of photosynthesis requires precise coordination between electron transport and CO2 assimilation, but how plants maintain this coordination under suboptimal CO2 and light conditions remains poorly understood. Here, we identify two tomato (Solanum lycopersicum) β-carbonic anhydrases (βCAs) responsive to defi...

Photosystem I structural fusion with IsiA enhances light-harvesting capabilities of filamentous cyanobacterium during iron deficiency

SUMMARY During iron stress, cyanobacteria produce iron-stress-inducible protein A (IsiA), which associates with photosystem I (PSI) to support cell growth. In most cyanobacteria, the stress induces formation of large IsiA18-PSI3 complexes. However, some species, including Anabaena sp. PCC 7120, studied in this work, fo...
