TL;DR

Overall, compound 15 represents a promising BCL6 PROTAC degrader for DLBCL, which induced rapid and efficient BCL6 degradation in OCI-LY1 cells and displayed potent antiproliferative activity in BCL6-dependent DLBCL cells.

Abstract

B-cell lymphoma 6 (BCL6) is a key transcriptional repressor involved in the pathogenesis of diffuse large B-cell lymphoma (DLBCL). In this study, scaffold hopping of BI-3802 led to compound 1, a new tricyclic BCL6-targeting ligand. Further optimization of linker architecture and CRBN-recruiting motifs led to compound 15, a potent BCL6 PROTAC degrader, which induced rapid and efficient BCL6 degradation in OCI-LY1 cells (DC50 = 5.32 nM) and displayed potent antiproliferative activity in BCL6-dependent DLBCL cells (IC50 = 3.2 nM). BCL6 degradation was CRBN- and proteasome-dependent and was accompanied by G1-phase arrest and increased p21 and p27 expression. Oral administration of 15 inhibited tumor growth in an OCI-LY1 xenograft model (TGI = 84% at 20 mg/kg) and reduced tumor BCL6 and Ki67 levels without appreciable body-weight loss or overt major-organ histopathology. Overall, compound 15 represents a promising BCL6 PROTAC degrader for DLBCL.

Discovery of CDK8/19 inhibitors as lead compounds for IL-6–dependent lymphoma enabled by the formal synthesis of cortistatin A

A formal synthesis of cortistatin A, a marine-derived steroidal alkaloid with potent anti-angiogenic and antileukemic activities, was achieved via a unique synthetic strategy that leveraged a late-stage SmI2-mediated oxa-bridge translocation. The de novo–synthesized simplified analog CA411 was evaluated across 38 cell...

The design, synthesis and antitumour evaluation of BCL3 inhibitors targeting advanced colorectal and breast cancers

The B-cell lymphoma 3 protein (BCL3) is a transcriptional facilitator protein and anti-cancer drug target driving intracellular cancer signalling pathways including NF-κB (advanced breast) and Wnt-β-catenin (colorectal). BCL3 governs pro-oncogenic processes such as tumour initiation, metastasis, and therapy resistance....

Discovery of Potent and Non-cytotoxic HDAC3-Selective Degraders Featuring a Propyl Hydrazide Zinc-Binding Group

Histone deacetylase 3 (HDAC3) is an important therapeutic target in cancer, yet selective HDAC3 degraders remain limited. In this study, we report the development of novel class I HDAC degraders derived from the HDAC3-preferential inhibitor FL-007. Two linker exit vectors were investigated, leading to the identificat...

Upamostat potentiates bortezomib cytotoxicity and delays proteasome recovery in bortezomib-resistant malignant lymphoma cells through modulation of the DDI2/NRF1 axis.

Findings identify UMS as a candidate DDI2/NRF1-axis modulator that potentiates BTZ-induced cytotoxicity and delays functional proteasome recovery in BTZ-resistant malignant lymphoma cells.

Discovery of new 7-azaindole derivatives as CSN5 inhibitors with in vitro antitumor activity against triple-negative breast cancer.

COP9 signalosome subunit 5 (CSN5) exerts critical deneddylase activity and is frequently overexpressed in triple-negative breast cancer (TNBC), representing a compelling antitumour target. However, to date, only a limited number of inhibitors targeting CSN5 have been reported, and none of them have advanced into clinic...

Discovery of 4-Aminoquinazoline Derivatives as Potent PKMYT1 Inhibitors via Hydrogen Bond Donor-To-Acceptor Conversion for CCNE1-Amplified Cancers.

PKMYT1 is a critical regulator of the cell cycle and has emerged as a promising synthetic lethal target for CCNE1-amplified cancers. In this study, we report the design, synthesis, and biological evaluation of a novel series of 4-aminoquinazoline-based PKMYT1 inhibitors. A hydrogen bond donor-to-acceptor conversion str...
