A near-infrared fluorescent probe based on an ICT mechanism that offers a reliable tool for studying oxidative stress in ALI and evaluating hepatoprotective agents, indicating that the probe reliably reports the severity of oxidative injury in ALI.

Abstract

Acute liver injury (ALI) is linked to oxidative stress, with hydrogen peroxide H2O2 as a key mediator. We report a near-infrared fluorescent probe, PhCy-H2O2, based on an ICT mechanism. The probe shows high sensitivity (detection limit in the low micromolar range) and excellent selectivity, producing a significant fluorescence turn-on at 796 nm upon H2O2 recognition. This reaction has the advantages of high sensitivity, high selectivity, fast reaction and stability under physiological pH conditions. In both cellular and mice models, the probe clearly visualized dose-dependent H2O2 generation induced by LPS and APAP, with fluorescence signals effectively attenuated by GSH and NAC, respectively, confirming the observed signals are associated with oxidative stress. Moreover, the hepatic fluorescence intensities correlated well with the levels of MDA and TNF-α, and the signal changes faithfully reflected the protective effects of antioxidant interventions, indicating that the probe reliably reports the severity of oxidative injury in ALI. Thus, PhCy-H2O2 offers a reliable tool for studying oxidative stress in ALI and evaluating hepatoprotective agents.

Development of a novel near-infrared fluorescent probe for nitroxyl imaging in acute kidney injury.

Nitroxyl (HNO), a short-lived reactive nitrogen species, participates in regulating cellular redox and nitrosative homeostasis, and its level fluctuations may reflect biochemical alterations associated with renal tubular injury during acute kidney injury (AKI). Therefore, the development of HNO fluorescent probes for A...

A dual‐channel fluorescent probe for in situ imaging of mitochondrial superoxide and microenvironmental changes in Cd‐induced hepatorenal dysfunction

Abstract Cadmium (Cd) is an environmental pollutant with high bioaccumulation and multi‐organ toxicity, posing a major threat to human health, especially to the liver and kidneys. Fluorescent probes have shown great potential for disease‐related imaging because of their high sensitivity and specificity. However, most e...

A near-infrared fluorescent probe for hypochlorous acid enables high-content screening of natural products for hypertension intervention.

Hypertension is accompanied by persistent oxidative stress and inflammatory activation, which contribute to vascular dysfunction and target-organ injury. Hypochlorous acid (HClO), a highly reactive oxidant generated through the myeloperoxidase pathway, may serve as a molecular indicator of hypertension-associated oxida...

Near-infrared fluorescence probe achieves the early diagnosis and efficacy assessment of pesticide-mediated liver injury by monitoring H2O2.

Recently, pesticide residues have become a serious environmental hazard due to the widespread employment of pesticides in agricultural production. With the continuous accumulation of pesticide in vivo, liver injury caused by pesticide metabolism has become a rapidly developing disease. Although pesticide-mediated liver...

Evaluation of a boronate-masked naphthalimide probe for peroxynitrite-responsive fluorescence imaging in experimental acute liver injury.

Peroxynitrite (ONOO-), a key mediator of oxidative and nitrosative stress, participates extensively in the pathological progression of acute liver injury and cancer. Because aberrant ONOO- production is closely associated with these conditions, accurate analysis of this short-lived species is important for early diagno...
