TL;DR

Combined treatment with the AXL inhibitor bemcentinib together with the RAS(ON) multi-selective tri-complex inhibitor RMC-7977 (RASi) or the ERK-selective inhibitor SCH772984 (ERKi) significantly enhanced growth suppression in human KRAS-mutant pancreatic and lung cancer models.

Targeting AXL Overcomes Adaptive Resistance to KRAS Inhibition in KRAS-Driven Cancer

By integrating adaptive response to KRAS inhibition with anti-tumor immunity, AXL represents a mechanistically actionable vulnerability whose inhibition deepens and prolongs responses to both allele-specific and pan-KRAS-targeted therapies.

Targeting Compensatory AKT Activation Overcomes Resistance to AXL Inhibition in Acute Lymphoblastic Leukemia.

The GAS6/AXL pathway plays a crucial role in promoting tumor survival and therapy resistance in various cancers. However, its significance in acute lymphoblastic leukemia (ALL) and the potential for its therapeutic targeting remain incompletely characterized. We integrated transcriptomic analysis of the public dataset...

XPO1 inhibition enhances the efficacy and durability of RAS-targeted therapy in preclinical models of KRASG12D mutant pancreatic ductal adenocarcinoma.

KRASG12D-selective and pan-RAS inhibitors have shown promise in pancreatic ductal adenocarcinoma (PDAC), yet adaptive resistance is anticipated to limit durability of response. Exportin 1 (XPO1), a nuclear export protein frequently overexpressed in PDAC, represents a potential vulnerability in KRAS-mutant cancers. We e...

Abstract B054: Combined RAS Inhibition and Immune Checkpoint Blockade Overcome NF-κB-Mediated Immune Evasion in Chemoresistant Pancreatic Cancer

Pancreatic ductal adenocarcinoma (PDAC) remains one of the most lethal malignancies. Surgery is the only potentially curative treatment, yet approximately 85% of patients present with unresectable disease. For these patients, chemotherapy remains the standard treatment, though nearly all develop resistance and diseas...

Abstract B068: FYN and other Src family kinases antagonize response to RAS inhibition in pancreatic ductal adenocarcinoma

Oncogenic KRAS drives pancreatic ductal adenocarcinoma (PDAC), yet resistance to RAS antagonists limit benefits. To define therapeutic response and resistance mechanisms, we deployed two complementary preclinical PDAC models, including a degron-regulated KRASG12D-dependent transplantation model, and a Kras G12V-drive...

Abstract B104: Pharmacological RAS inhibition synergizes with CAR T cells for tumor killing

Despite the success of chimeric antigen receptor (CAR) T cell therapy for liquid cancers, it has provided minimal clinical benefit in solid cancers, including pancreatic ductal adenocarcinoma (PDAC). We have previously shown that pharmacological RAS inhibition favorably remodels the PDAC tumor immune microenviron...
