The essential roles of heparan sulfate proteoglycans (HSPGs) in brain development, synaptic function, axonal guidance, synapse formation and stabilization, and the establishment of correctly interconnected neural networks are examined.

Abstract

ABSTRACT This narrative review examines the essential roles of heparan sulfate proteoglycans (HSPGs) in brain development, synaptic function, axonal guidance, synapse formation and stabilization, and the establishment of correctly interconnected neural networks. Distinct HSPG profiles help shape precise neural circuits, whereas HSPG dysregulation is associated with impaired neurodevelopmental processes, cognitive decline, and pathological changes in brain tissues. These findings emphasize the importance of HSPGs in normal brain function and in disease‐associated tissue dysfunction. HSPGs are a diverse group of proteins with critical roles in the CNS and PNS, including synaptic function, neurodevelopment, neurodegeneration, cell migration, and regulation of neuroprogenitor stem cell niches in the subventricular ependymal zone and subgranular dentate gyrus. Together, the diverse functions of HSPGs place them as master regulators of tissue form and function in health and disease.

A shared extracellular plasticity program drives brain repair and tumor progression

Injury induces complex multicellular repair responses that restore tissue integrity and replenish lost cells. In the brain, regeneration is limited and depends on the proliferation of neural and glial progenitors. But how these cells remodel the microenvironment to support repair remains poorly understood. Here, we pro...

Neuregulin-1 as a Context-Dependent Regulator of Neuroinflammation and Neural Repair: Mechanisms, Disease Relevance, and Therapeutic Challenges

Highlights What are the main findings? NRG1 emerges as a context-dependent regulator of neuroinflammation that coordinates microglial, astrocytic, oligodendroglial, neuronal, and neurovascular responses through ErbB receptor signaling rather than functioning as a uniformly anti-inflammatory molecule. Across multiple pr...

Nervous System Integrity in Drosophila: Essential and Dosage-Sensitive Roles of Hsp110.

The Hsp70 chaperone system is central in maintaining cellular proteostasis. In eukaryotes, its ATP/ADP nucleotide exchange cycle is stimulated by co-chaperone Hsp110, a divergent Hsp70 superfamily member and a modifier of protein misfolding disorders. Hsp110 chaperone functions and pathway interactions have been extens...

Multicellular Exosomes as Therapeutic Modulators of Neuroinflammation: Mechanisms and Clinical Prospects

Abstract Neuroinflammation is a fundamental pathological driver of diverse central nervous system (CNS) disorders, including Alzheimer’s disease, stroke, and traumatic brain injury. This complex process is characterized by persistent neuronal damage and the orchestrated response of glial and peripheral immune cells. In...

Fibroblasts as key regulators of neuro-mesenchymal niches in homeostasis, inflammation and disease

Fibroblasts, once viewed as inert producers of extracellular matrix (ECM), are now recognized as dynamic and heterogeneous regulators of tissue identity, homeostasis, and repair. Single-cell and spatial transcriptomic studies have revealed extensive transcriptional and positional diversity among fibroblast population...
