Same hydrolase knockouts, different outcomes: CRISPR/Cas9 editing workflow shapes resulting cellular phenotype and transcriptome in CHO

CRISPR/Cas9-based multi-gene knockout strategies are widely applied to engineer Chinese hamster ovary (CHO) cells, yet the impact of editing workflow on cellular outcomes remains poorly understood. Here, we systematically compare sequential and multiplex genome editing approaches to generate CHO cell lines lacking nine...

Expanding Transposase Technology to Regulatory Noncoding RNAs for Stable Expression in CHO Cells

A transposase‐mediated platform for stable microRNA (miRNA) expression in CHO cells is established and the first application of a DNA transposon system for long‐term miRNA genome integration is demonstrated, demonstrating the applicability of transposase systems beyond protein‐coding transgenes.

Engineering Stable Lentiviral Vector Producer Cells Improves LVV Production.

The results demonstrate that transcriptomics-guided host-cell engineering is an effective strategy to identify and relieve intrinsic bottlenecks in stable LVV producer cell lines.
