TL;DR

Targeting the FTO-GPX4 axis may offer novel therapeutic strategies for ALI, and silencing FTO in vivo significantly ameliorates pulmonary tissue pathology, impaired lung function, ferroptosis, and inflammatory responses in mice, whereas GPX4 inhibition reverses this protective effect.

Sufentanil attenuates LPS-induced acute lung injury by suppressing ferroptosis and maintaining GPX4 protein abundance

Introduction Acute lung injury (ALI) is a severe inflammatory syndrome characterized by disruption of the alveolar-capillary barrier, pulmonary edema, and impaired gas exchange. Ferroptosis has emerged as an important contributor to ALI pathogenesis, but the mechanisms associated with reduced glutathione peroxidase 4 (...

Acacetin alleviates sepsis-induced diaphragmatic dysfunction by inhibiting ferroptosis via the PI3K/AKT/NRF2 pathway

The mechanism underlying the protective effects of Acacetin (AC) against sepsis-induced diaphragmatic dysfunction (SIDD) remains to be elucidated. AC is present in several medicinal plants and has many pharmacological effects and has been reported to inhibit ferroptosis. This study aimed to investigate the role of AC...

Itaconate alleviates thioacetamide-induced acute liver injury by suppressing pyroptosis-associated mitochondrial dysfunction and activating the Nrf2/HO-1 pathway

Findings indicate that 4-OI alleviates TAA-induced acute liver injury, at least in part, through activation of the Nrf2/HO-1 signaling pathway and suppression of pyroptotic activation and mitochondrial dysfunction-associated inflammatory amplification.

FOXA1-driven transcription and TRIM9-mediated ubiquitination of LCN2 modulate ferroptosis in experimental sepsis-induced acute lung injury

Sepsis-induced acute lung injury (ALI) remains a devastating complication of sepsis, marked by uncontrolled pulmonary inflammation, alveolar-capillary barrier disruption, and high mortality. Ferroptosis, an iron-dependent form of regulated cell death driven by lipid peroxidation, is implicated in sepsis-related org...

FTY720 attenuates LPS-induced acute lung injury through PP2A- tristetraprolin–dependent anti-inflammatory signaling

Acute lung injury (ALI) is characterized excessive inflammatory signaling and uncontrolled cytokine production. Tristetraprolin (TTP) is a key post-transcriptional regulator of inflammatory cytokines that negatively regulates inflammatory pathways. PP2A emerges as an important player to activate TTP. However, wheth...
