NMDA receptor hypofunction on GABAergic interneurons results in input-specific Excitatory/Inhibitory imbalance in pyramidal neurons of medial prefrontal cortex
It is found that circuit dysfunction can selectively bias the processing of specific afferent pathways rather than produce a uniform disruption of cortical excitability, with direct relevance for understanding hippocampal–prefrontal dysconnectivity in neuropsychiatric disorders.
It is discovered that d-serine but not glycine is critical for maintaining the activity of NMDARs at FS-PV+ interneurons in the prelimbic area of the prefrontal cortex and that loss of its functions would recapitulate the synaptic deficits observed in several neuropsychiatric diseases like schizophrenia.

It is proposed that the simultaneous connections onto LC‐NA neurons and LC‐iINs, along with cell type–specific differences in transmitter release plasticity, may enable the mPFC to effectively multiplex information to the LC for adaptive behavioral regulation.

Basal forebrain cholinergic neurons regulate cortical activity and cognition and are vulnerable in Alzheimer's disease (AD). However, the upstream circuits controlling projection-defined basal forebrain cholinergic populations remain incompletely understood. Here, we used projection-specific rabies-mediated monosynapti...

Overall, aging differentially disrupts excitability and ionic mechanisms in pyramidal and interneuron populations, leading to impaired synaptic integration and altered short-term plasticity in mPFC microcircuits, suggesting that disrupted synaptic integration and intrinsic neuronal function in the aged neocortex may co...

PIC GABAergic neurons receive dense inputs from the isocortex and thalamus and send prominent projections to the basal ganglia and amygdala, with these brain regions constituting key hubs of the whole‐brain connectivity network.

This work uses cell type-specific retrograde anatomy and ex vivo slice physiology and optogenetics to characterize the properties of synapses onto CINs in the NAc core of mice of either sex, and establishes how these inputs and receptors evoke action potentials and influence spontaneous firing.
