TL;DR

This work successfully transformed UTEX 3222 by natural transformation, generating a large panel of fully segregated axenic strains and establishes the foundation for developing UTEX 3222 as a scalable platform for photosynthetic biomanufacturing.

Abstract

Cyanobacterium aponinum UTEX 3222 is a fast-growing marine cyanobacterium that reaches high cell densities, making it an attractive chassis strain for low-cost biomanufacturing. However, past efforts to genetically engineer UTEX 3222 have been unsuccessful. We hypothesized that the complex restriction modification systems in the UTEX 3222 genome constitute a major barrier to genetic transformation. Using a combined methylation avoidance and methylation mimicking approach, we overcame this barrier and successfully transformed UTEX 3222 by natural transformation, generating a large panel of fully segregated axenic strains. Transformation method was also established for a novel shuttle vector that replicates in UTEX 3222. A large-scale promoter screen identified promoters which exhibited strong expression at high cell density. These advances establish the foundation for developing UTEX 3222 as a scalable platform for photosynthetic biomanufacturing.

Identification of genetic determinants that promote biofilm growth under heterotrophic conditions in Cupriavidus necator using transposon enrichment

Results identify B2043 as a negative regulator of biofilm formation acting predominantly during attachment, provide the first experimental evidence for c-di-GMP-dependent biofilm regulation in C. necator H16, and establish a functional-genomics framework — together with eleven further candidate loci — for engineering p...

Development of a genetic toolkit for Sphingomonas and its application in tailored welan gum fermentation from bagasse hydrolysate.

Sphingomonas represent a significant microbial resource with considerable industrial applicability. However, non-model microorganisms often lack efficient genetic modification tools, which hinders the development of microbial cell factories. In this study, using the welan gum-producing strain Sphingomonas sp. HT-1 as a...

Version 2.0 of the Hermes transposon-based mutagenesis system for prototrophic mutant pool generation in Candida glabrata

ABSTRACT Although the original Hermes transposon mutagenesis system has proven successful for saturated insertional mutagenesis in Candida glabrata, its reliance on uracil auxotrophy may limit its applicability under nutrient-restricted host environments. To overcome this limitation, Zaeske et al. (Microbiol Spectr 14:...

Genetic engineering of cyanophages for harmful cyanobacterial bloom treatment—A mini review

Harmful cyanobacterial blooms (HCBs) are a prominent worldwide environmental concern. Proliferation of HCBs outpaces the development of mitigation and management technologies. Recently, cyanophages began to emerge as a potential abundant resource that can be harnessed for target-specific HCB treatment. However, there e...
