Insight is given into the genetic regulatory networks within the HPG axis that orchestrate testicular development and maturation in roosters, paving the way for potential applications in the molecular breeding of breeding roosters for reproductive optimisation.

Abstract

1. Despite the pivotal role of the hypothalamic-pituitary-gonadal (HPG) axis in avian reproduction, the underlying molecular mechanisms that govern sexual maturity and spermatogenesis in roosters are not well elucidated. This study aimed to explore gene expression changes in the HPG axis using Wenchang cockerels.2. Phenotypic results showed significant testicular growth from 18 to 20 weeks of age. Transcriptome analyses identified 2,154 differentially expressed genes (DEG) in testes, which was markedly higher than those identified in hypothalamus (52 DEG) and pituitary (27 DEG).3. Functional enrichment analysis showed neuroactive ligand-receptor interactions and hormone transport pathways in the hypothalamus and pituitary and ciliary motility and spermatogenesis pathways in the testes.4. Protein-protein interaction (PPI) networks analysis pinpointed key hub genes, such as BDNF, NPTX2, TEKT2, TEKT3, CFAP52, and MNS1, associated with spermatogenesis and sperm motility.5. This research gave insights into the genetic regulatory networks within the HPG axis that orchestrate testicular development and maturation in roosters, paving the way for potential applications in the molecular breeding of breeding roosters for reproductive optimisation.

Integrated 4D-DIA proteomic and transcriptomic profiling delineates the molecular trajectory of yak testicular development

Normal testicular development is essential for sustaining male fertility in livestock, yet the underlying molecular regulatory mechanisms remain poorly understood, particularly in environmentally adapted breeds such as the yak. In this study, we employed an integrated 4D-DIA proteomic and transcriptomic strategy...

Early gonadal development and effects of 17α-methyltestosterone-induced sex reversal on physiological parameters and gonadal gene expression in snakeskin gourami.

The plasticity period of sexual development facilitates sex manipulation in fish through exogenous hormone treatment. However, whether 17α-methyltestosterone (MT)-treated males exhibit growth performance and physiological traits similar to those of naturally genotyped males remains unclear. We evaluated MT treatment ef...

The Transcription Factor PRDM15 Activates the PPP2R5C/PI3K‐Akt Pathway to Promote Steroid Hormone Secretion in Goat Granulosa Cells

The proliferation and steroid hormone production capacity of granulosa cells (GCs) are critical factors in determining follicular maturation and female reproductive success. This study aims to investigate the role of PPP2R5C in regulating GC function and to reveal its upstream transcriptional regulatory mechanisms, the...

139. Award Talk: Integrative Physiology and Functional Genomics Reveal Novel Mechanisms Regulating Pubertal Development in Gilts.

Age at puberty is the earliest indicator of future reproductive potential of a gilt and because it is moderately heritable, identification of genomic markers can support marker-assisted selection to improve gilt development.
