Overall, treatment selection is increasingly determined by tumour type, stage, resectability and validated biomarkers, while several important questions concerning optimal sequencing, long-term outcomes and under-represented patient groups remain unresolved.
Abstract
Simple Summary Cutaneous malignancies include melanoma, cutaneous squamous-cell carcinoma, basal-cell carcinoma and Merkel-cell carcinoma. Their management has changed rapidly in recent years owing to the increasing use of immune checkpoint inhibitors, targeted therapies and perioperative systemic treatment. This review summarizes the current multimodal treatment landscape, including surgery, radiotherapy, immunotherapy, molecularly targeted therapy and selected reconstructive strategies. Particular attention is given to recent practice-changing evidence, including neoadjuvant immunotherapy in resectable stage III melanoma, adjuvant cemiplimab in high-risk cutaneous squamous-cell carcinoma, anti-LAG-3 therapy in advanced melanoma and Hedgehog-pathway inhibition in advanced basal-cell carcinoma. The review also discusses predictive biomarkers, treatment sequencing, resistance mechanisms and special populations such as transplant recipients. Overall, treatment selection is increasingly determined by tumour type, stage, resectability and validated biomarkers, while several important questions concerning optimal sequencing, long-term outcomes and under-represented patient groups remain unresolved.
BACKGROUND
The prognosis of metastatic nasopharyngeal carcinoma (mNPC) remains dismal despite recent advances of locoregional and systemic therapy. The incorporation of programmed death-1 (PD-1) inhibitors into first-line chemotherapy has transformed the therapeutic landscape; however, optimal treatment strategies beyo...

Melanoma is a highly aggressive skin cancer with a steadily increasing annual incidence. Although it accounts for only 1% of diagnosed skin cancers, it has the highest mortality rate among them. Therefore, it is essential to continuously develop treatment methods and improve long-term outcomes. In recent years, a break...

Outcomes for patients with resectable head and neck squamous cell carcinoma (HNSCC) remain suboptimal despite multimodal therapy, with high rates of recurrence and patients suffering from toxicity of treatments. Immune checkpoint inhibitors, which have transformed the management of recurrent and metastatic disease, hav...

Simple Summary Cutaneous squamous cell carcinoma is a common type of skin cancer that is usually cured with surgery. However, some patients develop advanced disease that cannot be removed surgically or spreads to other parts of the body. Although immunotherapy has improved outcomes, many patients do not respond to trea...
Overall, TIL therapy constitutes a rapidly evolving and clinically potent treatment modality for melanoma and covers recent advances for improving therapeutic outcomes, including safety optimization, product optimization, next-generation engineered TILs, and predictive biomarkers.
