Genetic evaluation of encorafenib-associated kinases RAF1 and MAPK8 in pancreatic cancer: Exploratory mediation by circulating immune traits

Highlights • Genetically proxied RAF1 and MAPK8 expression was associated with pancreatic cancer risk.• A circulating CD4+ Treg-related phenotype showed a possible partial indirect contribution.• PheWAS detected no broad genetic pleiotropic signals at the prespecified threshold.• The findings support further study of RAF1- and MAPK8-related pathways in pancreatic cancer.
