TL;DR

Cryo-electron microscopy structures of the Arabidopsis thaliana ammonium transporter AMT1;1 in apo, ammonium-bound, methylammonium-bound, low-pH and phosphomimetic states reveal a conserved ammonium conduction pathway comprising an aromatic recruitment vestibule, a Phe-gate/Twin-His-containing pore, and a polar intracellular exit region.

Abstract

Efficient ammonium uptake is essential for plant growth and nitrogen-use efficiency, yet the structural basis of ammonium transport and regulation in plant AMTs remains unclear. Here, we determined multiple cryo-electron microscopy structures of the Arabidopsis thaliana ammonium transporter AMT1;1 in apo, ammonium-bound, methylammonium-bound, low-pH and phosphomimetic states. These structures reveal a conserved ammonium conduction pathway comprising an aromatic recruitment vestibule, a Phe-gate/Twin-His-containing pore, and a polar intracellular exit region. We further identified a conserved extracellular N-terminal disulfide bond required for oligomer stability and transport activity, suggesting potential redox-dependent regulation. On the cytoplasmic side, phosphorylation modulates C-terminal interactions with intracellular loops, revealing an allosteric regulatory mechanism. Comparison with the bacterial ammonium-sensing protein Sd -Amt1 uncovers a conserved ammonium-recognition region in At AMT1;1 that contributes to transport activity but lacks the signaling module and ligand-dependent conformational switching observed in Sd -Amt1. Together, these findings provide structural insight into ammonium transport and coordinated regulation in plant AMT transporters.

Structural basis for phosphorylation-induced inactivation of the rice ammonium transporter AMT1;1

Ammonium (NH4+) uptake and transport in plants are mediated by ammonium transporters (AMTs), which are tightly regulated by phosphorylation to prevent excess ammonium influx and toxic accumulation. However, the mechanism of phosphorylation-mediated inactivation is unique and remains unclear. Here, we report cryo-EM str...

Lipid-associated architecture and divergent ammonium transport distinguish a bacterial Rhesus protein

Nitrification is a major process in the global nitrogen cycle, initiated by the oxidation of ammonia to nitrite. The ammonia-oxidising bacterium Nitrosomonas europaea catalyses this first and rate-limiting step and depends on ammonium both as an energy substrate and as a nitrogen source, making ammonium acquisition cen...

Membrane environment-dependent relocation of phloem-loading sucrose transporters.

Plasma membrane transporters play an essential role in plant development and all physiological processes. Their activity is tightly controlled by a variety of mechanisms, including the rate of transporter endocytosis. There is evidence that the local membrane environment, in particular the sterol content, influences tr...

NRT1.1 as central signaling hub orchestrating plant growth and stress trade-offs

Plants must continuously balance nitrogen-dependent growth with adaptation to environmental stresses. The nitrate transporter AtNRT1.1 in Arabidopsis thaliana and its rice ortholog OsNRT1.1B have been reported to integrate nitrogen- and stress-related signals in their respective species. This review synthesizes how NRT...

CAX1 beyond calcium transport: evidence for a transport-independent regulatory module in plants

Plant cation/H+ exchanger 1 (CAX1) is a vacuolar Ca2+/H+ antiporter that plays a central role in calcium homeostasis. The initial 36 residues of CAX1 make up the amino-terminal regulatory region (NRR), this defining feature mediates autoinhibition and serves as a target for phosphorylation and protein-protein interacti...

A bacterial Rhesus transporter retunes a structurally conserved ammonium pore into a reversible nitrogen valve

How conserved proteins acquire new physiological functions is a central question in molecular evolution. Rather than inventing new architectures, evolution often repurposes existing scaffolds, preserving core structural features while retuning the molecular logic that connects mechanism to physiology. Membrane transpor...
