Effect of Different Temperature, pH, Carbon and Nitrogen Sources on Growth of Rhizoctonia solani in vitro
Abstract
Fenugreek (Trigonella foenum-graecum L.) is an annual forage legume and spice crop. It has anti-diabetic, anti-cancerous, anti-microbial and hypocholesterolaemic properties. This crop is attacked by many of fungal and bacterial diseases although root rot disease (Rhizoctonia solani Kuhn) is one of most destructive among them. This disease greatly favoured by high temperature and negative with rainfall and in sandy soil of Rajasthan, where the climatic conditions are very adverse, due to dry and high temperature. Therefore, it was very important to find out the physiological parameters of Rhizoctonia solani for disease control. The present study concluded with, in different temperature highest mycelial growth was recorded at 30°C followed by 25°C, 35°C, 20°C and found least at 15°C. In case of pH highest mycelial growth was observed at pH 7.0 followed by pH 7.5, pH 6.5, pH 8.0 and found minimum at pH 8.5. In carbon sources highest mycelial growth was recorded in sucrose followed by glucose, maltose, fructose and lactose. However, in nitrogen sources highest mycelial growth was observed in L-alanine followed by glutamic acid, ammonium nitrate and L- arginine. However, ammonium chloride yielded the lowest mycelial growth.
