Abstract

To understand neural computations, one would like to relate circuit dynamics to neuronal intrinsic excitability and connectivity. We developed tools for large-scale all-optical electrophysiology and applied them to approximately 300 layer 1 interneurons per mouse in vivo. Across two mice, targeted stimulation and voltage imaging identified 1,469 inhibitory and 5,752 directed electrical connections among 14,312 tested pairs. Presynaptic identity was more strongly associated with inhibitory post-synaptic potential (IPSP) amplitude, whereas postsynaptic identity was more strongly associated with IPSP decay time. Gap junctional and synaptic interactions had distinct spatiotemporal kernels, with corresponding signatures in spontaneous activity. Excitability measured under optogenetic stimulus predicted spiking responses to subthreshold voltage, depolarization rate, and spike history during spontaneous firing. Genetic subtypes also showed different behavioral-state-dependent voltage-to-spike relationships despite similar population-average subthreshold voltages. These measurements link cellular properties and pairwise interactions to spontaneous circuit dynamics, establishing an approach for functional circuit mapping in vivo. Highlights All-optical mapping links excitability, connectivity, and spontaneous dynamics in vivo Presynaptic and postsynaptic identity relate differently to IPSP strength and decay Gap junctional and synaptic couplings reveal distinct spatiotemporal interactions Measured intrinsic excitability predicts features of spontaneous spiking in vivo

Excitability across scales: a unifying EI model

Neuronal excitability is commonly modeled using different principles at the cellular and population scales, making it difficult to study how microscopic mechanisms shape macroscopic activity. We introduce a spatiotemporal excitatory-inhibitory (EI) model that unifies these scales through a common mixed-feedback archite...

Photomapping electrically coupled networks of the mature thalamus and cortex

Electrical synapses are expressed ubiquitously across the brain and are crucial components of active neural circuitry and connectomes. Identification of coupled networks in living tissue is limited by technical demands of multiplexed recordings, and no dyes, fluorescent reporters, or genetic labels can now fill the gap...

Learning induces activation-mechanism–dependent neural plasticity in an intracortical microstimulation task

Results show that learning in an ICMS task reshapes cortical circuits through activation-mechanism–dependent plasticity, underscoring the need for stimulation paradigms that adapt to both cell-intrinsic and network dynamics.

Sensory experience drives glutamate spillover, presynaptic LTP and functional connectome remodeling in the barrel cortex

Neural-network simulations suggest that volume-transmitted excitatory signals can improve associative memory retrieval in sparsely connected networks and point to a potential computational consequence of glutamate spillover for brain circuit function.

Granger sensori-behavioral functional taxonomy of neuronal ensemble activity from two-photon calcium imaging data

Understanding how neuronal populations interact to encode and transform sensory information is a fundamental challenge in computational neuroscience. Most existing studies, however, study neural encoding, behavioral readout, and functional connectivity as disjoint problems. Two-photon calcium imaging enables simultaneo...

Computational Insights Into Neuromodulation: How Electric Fields Shape Motor Neuron Excitability

It is suggested that low-intensity EEFs produce modest, morphology-dependent modulation of MN electrophysiological properties, and that variability in neuronal structure and orientation may help explain discrepancies across previous experimental and modeling studies.
