Vitronectin-driven amyloid deposition disrupts mitochondrial homeostasis via integrin-mediated pathway in a humanized mouse model of transthyretin cardiac amyloidosis.
Modulation of integrin signalling represents a potential complementary therapeutic strategy in ATTR-CM beyond TTR suppression and actively drives cardiac amyloid deposition, and disruption of integrin αvβ3/FAK/Akt signalling links amyloid-ECM interactions to mitochondrial dysfunction.
These findings identify the PGAM5/VDAC1 axis as a key mechanism linking ponatinib stress to coordinated failure of MQC in the heart and targeting PGAM5-dependent VDAC1 oligomerization may represent a potential strategy for limiting ponatinib-associated cardiotoxicity.
Cardiomyocyte-targeted AAV9 delivery of phospho-mimic âIII-tubulin improves Cx43 organization, inflammatory remodeling, and arrhythmia susceptibility in dystrophic hearts, demonstrating that therapeutic modulation of βIII-tubulin Ser172 phosphorylation partially recapitulates the protective effects observed in the gene...

A central hallmark of Parkinson's disease (PD) is the spread of -Synuclein (-Syn) aggregates, which is thought to contribute to its progressive nature. To better understand the mechanism of cellular internalization of -Syn fibrils, we conducted a genome wide CRISPR activation (CRISPRa) screen to identify genetic modifi...

This work describes LW-1017, a potent, selective, and brain-penetrant small-molecule TRPML1 agonist and evaluates its pharmacodynamic activity and therapeutic effects in aged preclinical mouse models of Parkinson’s disease (PD) and Alzheimer’s disease (AD), supporting TRPML1 agonism as a promising strategy to counterac...

Doxorubicin is a widely used chemotherapeutic agent whose clinical utility is limited by dose-dependent cardiotoxicity. The precise mechanisms bridging mitochondrial dysfunction to cardiac inflammatory responses remain poorly defined.
We utilized cardiomyocyte-specific CMPK2 knockout and overexpression mou...
