These findings elucidate key miRNA-mediated regulatory modules governing ToMMV resistance, advancing the understanding of host defense mechanisms against tobamoviruses.

Abstract

Background

Tomato mottle mosaic virus (ToMMV) is an emerging tobamovirus that posing a significant threat to the yield and quality of solanaceous crops globally. To elucidate the regulatory role of small RNAs in the host response to ToMMV, we used Nicotiana tabacum var. Xanthi nc as a model system and performed small RNA sequencing across three distinct disease stages: mosaic symptom onset, linear leaf symptom development, and the recovery phase. By integrating qRT-PCR validation with functional assays, including miRNA overexpression, virus-induced gene silencing, and biological inoculation, several miRNAs specifically associated with ToMMV resistance were identified.

Results

Functional characterization revealed that overexpression of miR171c, miR172a, and a novel miRNA (novel-miR363) significantly suppressed ToMMV accumulation and thereby enhancing plant resistance, whereas silencing of miR171c resulted in an approximately threefold increase in viral accumulation. In contrast, overexpression of another novel miRNA, novel-miR1022, promoted viral accumulation and facilitated viral infection. Further analysis through target prediction and experimental validation confirmed that miR171c and miR172a target transcripts encoding Scarecrow-like (SCL) and APETALA2 (AP2) class transcription factors, respectively. Specifically, miR171c appears to confer resistance by repressing NtSCL6 and NtSCL22, whereas miR172a likely mediates this effect through the downregulation of NtRAP2-7.

Conclusion

These findings elucidate key miRNA-mediated regulatory modules governing ToMMV resistance, advancing the understanding of host defense mechanisms against tobamoviruses.

Plant-mediated RNAi via Panonychus citri-derived miRNAs disrupts adult emergence and confers mite resistance.

MicroRNAs (miRNAs), small non-coding RNAs that trigger RNA interference (RNAi), are promising candidates for pest control, yet their efficacy against non-insect pests remains poorly explored. This study evaluates endogenous mite-derived miRNAs as transgenes for crop resistance against Panonychus citri, a major pest wit...

The OsAGO2-miR11337-5p-OsLRR Module Confers Rice Resistance to Rhizoctonia solani by Modulating Vitamin B6 Biosynthesis.

Argonaute (AGO) proteins play a pivotal role in small RNA (sRNA)-mediated RNA silencing and plant immunity. However, their roles in rice defense against sheath blight remain largely unknown. In this study, we found that the osago2 mutant exhibited enhanced resistance to Rhizoctonia solani (R. solani). Small RNA sequenc...

The miR168a-TaAGO1b module improves Fusarium head blight resistance and yield in wheat.

Fusarium head blight (FHB) is a devastating fungal disease of wheat that causes severe yield loss and mycotoxin contamination. Wheat varieties offering higher grain yield while maintaining moderate resistance to FHB are highly desirable. Here, we report that microRNA168a (miR168a) acts as a key negative regulator of bo...

A Csi‐miR3954b‐CsCE70/CsPLA1 Regulatory Module Enhances Citrus Fruit Resistance to Penicillium digitatum by Promoting Phenolic Acid and Flavonoid Biosynthesis

ABSTRACT Green mould is one of the major postharvest diseases affecting citrus. Although evidence indicates that microRNAs (miRNAs) participate in plant defence responses, their roles and regulatory mechanisms in citrus resistance to green mould remain unclear. Small RNA sequencing identified several defence‐associated...
