Investigation of the safety and therapeutic potential of (2E,5E)-2,5-bis(isoquinolin-5-ylmethylene)cyclopentanone (ICP) against Leishmania infantum indicates that it represents an interesting chemical scaffold for further optimization and investigation toward the development of new treatments for leishmaniasis.
Abstract
Leishmaniasis is a neglected tropical disease whose current treatments present several limitations. In this context, the search for new antileishmanial agents remains highly relevant. The aim of this study was to investigate the safety and therapeutic potential of (2E,5E)-2,5-bis(isoquinolin-5-ylmethylene)cyclopentanone (ICP), a bis(arylmethylidene) cycloalkanone, against Leishmania infantum. The antileishmanial activity of ICP was evaluated against intracellular amastigote forms of L. infantum, and the compound exhibited an EC₅₀ value of 22.6 ± 0.17 μM. The cytotoxicity of ICP was also assessed in the human monocytic leukemia cell line THP-1, revealing low toxicity, with a CC₅₀ of 190 ± 2.5 µM and a selectivity index of 8.4. In addition, in vivo studies were performed to investigate acute toxicity and therapeutic efficacy. For the evaluation of acute toxicity in healthy mice, ICP was administered intraperitoneally at a single dose of 100 mg/kg. Under these conditions, no deaths or body weight loss were observed. Moreover, no significant changes were detected in hematological, hepatic, cardiac, or renal parameters in ICP-treated animals when compared with the control group. Therapeutic efficacy was evaluated in L. infantum- infected mice treated intraperitoneally with ICP at doses of 25, 50, and 75 mg/kg/day for 10 consecutive days. A reduction in parasite load (85%) was observed in the liver of treated animals compared with the control group. Overall, these preliminary results indicate that ICP represents an interesting chemical scaffold for further optimization and investigation toward the development of new treatments for leishmaniasis.
The molecular modeling findings suggest a potential interaction between E9 and trypanothione reductase, supporting the need for further evaluation of these compounds in intracellular amastigote models and subsequent in vivo studies and suggesting that the biological activity of this compound series may differ among Lei...
The urgent need for new antileishmanial drugs persists due to resistance and toxicity associated with current treatments. Although bisindoles represent a promising chemotype, our first-generation 2,2'-di(indol-3-yl)ethanamines, inspired by a natural marine alkaloid, displayed significant cytotoxicity despite potent act...

The TA derivatives demonstrated a more favorable balance between antiproliferative potency, tumor selectivity, and effects on wound-closure than the corresponding TAFe complexes, although selected TAFe derivatives exhibited cell line-dependent selectivity.

Among 29 compounds, tetraoxanes 27, 29, 30, 32, and 33 exhibited the highest potency against both Leishmania species, but most showed limited selectivity toward the parasite, whereas 33 demonstrated strong efficacy against L. major.

A new series of compounds was evaluated as prospective in vitro antimalarial agents against the chloroquine‐sensitive 3D7 and chloroquine‐resistant K1 strains of Plasmodium falciparum, and the compound bearing a 4‐chloroaryl moiety was the most active against P. falciparum.

The preparation of a new class of organochalcogen compounds derived from arylchalcogeno aminothymidine (ACAT) and 2,4,6-trichloro-1,3,5-triazine is described. The compounds were obtained in good yields via aromatic nucleophilic substitution reactions between ACAT derivatives and the triazine ring. The developed proto...
