TL;DR

Findings indicate that phage propagation in Omptin-expressing bacteria leads to the loss of dibasic amino acid motifs from phage-displayed combinatorial peptide, protein, and antibody libraries, which suggests that the presence of dibasic residues in proteins and peptides displayed on bacteriophage M13 should necessitate the use of OmpT- and OmpP-deficient bacterial hosts.

Sequence Similarity Network-Guided Discovery of a Sequence-Tolerant Cyanobactin Protease Clade for Site-Selective Peptide and Protein Modification

Proteases responsible for leader peptide removal are ubiquitous in ribosomally synthesized and post-translationally modified peptide (RiPP) biosynthetic pathways. However, many characterized RiPP proteases require specific core peptide modifications prior to cleavage, limiting their broader applicability. Here, we id...

A generalizable method for the purification and biophysical characterization of bacterial membrane receptors

Abstract Membrane‐embedded bacterial receptors are challenging to express and purify in soluble form, yet their isolated domains are essential tools for structural and ligand‐discovery studies. Pseudomonas aeruginosa relies on the TonB‐dependent heme receptor HasR for iron acquisition, a process central to its pathogen...

Structural and Functional Interrogation of Active Streptococcus pneumoniae Sortase A

Sortase A (SrtA) enzymes covalently anchor surface proteins to Gram-positive bacterial cell walls, promoting colonization and virulence. In Streptococcus pneumoniae, previous studies identified both a domain-swapped dimer and an active refolded monomer, but the active enzyme has not been characterized at the structural...

Structural insights into the polytopic Escherichia coli phosphoglycosyl transferase WbaP.

The 3.0 Å cryo-electron microscopy apo-structure of the PGT enzyme WbaP from Escherichia coli, a UDP-galactose:undecaprenolphosphate galactose-1-phosphoryl transferase, provides new insights into the PGT family of enzymes.

[Screening of highly active recombinant bovine enterokinase mutants mediated by green fluorescent protein and its soluble expression in Escherichia coli].

The soluble expression of EKL in E. coli is achieved and a highly active mutant is obtained, and a visual tool for the rapid detection of EKL activity is provided.
