These data provide a configuration for driving nuclear and secreted proteins from a single-copy transgene and indicate that downstream secreted proteins could be correctly directed to the secretory pathway.
Abstract
Multicistronic expression systems enable production of multiple proteins from a single transcript, with internal ribosome entry sites (IRES), SL2 trans-splicing, and 2A peptides as common tools. Because 2A peptides rely on a single translation event, we tested whether nuclear-localized mStayGold (mSG::H2B) and secreted mScarlet (ssmScarlet) reporters separated by F2A in C. elegans produced the expected localization pattern. mSG::H2B::F2A::ssmScarlet and ssmScarlet::F2A::mSG::H2B expressed in body wall muscle produced nuclear mSG in muscle and ssmScarlet in coelomocytes, indicating that downstream secreted proteins could be correctly directed to the secretory pathway. These data provide a configuration for driving nuclear and secreted proteins from a single-copy transgene.
Messenger RNA (mRNA) localization to specific subcellular regions is crucial for proper protein translation and localization. Potential means to localize mRNAs include cis-acting or structural motifs embedded within the sequence. We previously identified a secretion-enhancing cis regulatory targeting element (SECReTE)...

It is proposed that MraW modification of 16S rRNA enhances translation efficiency in general, and that specific transcripts have evolved structural features that fine-tune protein levels that may be prevalent in bacteria which exhibit uncoupled transcription and translation.

Cryo-electron tomography of native and antibiotic-perturbed Mycoplasma pneumoniae cells is used to resolve 140 maps that recapitulate bacterial translation during the initiation, elongation, and recycling phases, allowing us to propose a threading-based translation reinitiation mechanism and to provide structural evide...

The cytosolic ATP-binding cassette F (ABC-F) family modulates ribosomal activity in eukaryotes and prokaryotes. Although they all bind to the E site of the ribosome, some bacterial ABC-Fs confer resistance to ribosome-targeting antibiotics, while housekeeping members have poorly defined roles. Interestingly, in Escheri...

Ice-binding proteins (IBPs) are functional molecules that enable organisms to survive severe cold by binding specifically to ice crystals and suppressing their growth and recrystallisation. Although their primary role has traditionally been viewed as antifreeze activity, recent studies have suggested additional cell-pr...

Transcriptional shut-off experiments confirmed that m6A has opposing effects on mRNA decay depending on cellular context: it stabilizes transcripts in differentiated cells, while it promotes the degradation of mRNAs associated with neoblasts.
