Abstract

The El-Rakhawi Pre-Clinical Photonic-Bio Verification System for CRISPR (DOI: 10.5281/zenodo.23507507) addresses the critical safety bottleneck in CRISPR gene editing: off-target effects that can cause oncogenic mutations. This PCT-ready patent specification discloses a revolutionary lab-on-a-chip architecture replacing conventional electronic simulation with photonic computing at the speed of light. The system integrates three core innovations: (1) a nanophotonic channel array with zwitterionic-PEG coating and 37°C thermal regulation preserving Cas9 RNP integrity; (2) a photonic processor utilizing Mach-Zehnder interferometers and microring resonators for optical matrix multiplications with O(1) propagation time, bypassing O(L^2) electronic bottlenecks; (3) a convolutional neural network specifically configured to map physical optical interference patterns to binding classifications, overcoming abstract idea rejections. Validated on benchmark datasets, the system achieves up to 99.99% sensitivity and 99.95% specificity, reducing validation time from weeks to under 30 minutes with 1,200 samples/day throughput. Comprehensive IP protection covers 15 claims spanning system, method, chip, and kit, with explicit prohibitions against unauthorized commercialization or AI training ingestion. All moral, financial, and proprietary rights exclusively reserved to Dr. Mohamed Kamal Arafa El-Rakhawi—establishing a commercially viable, regulatory-ready pathway for safe personalized gene therapy worldwide.
