Dual MEK and mTORC1/2 blockade resulted in synergistic antiproliferative effects in cell lines bearing alterations in KRAS/BRAF and PIK3CA/PTEN and a marked proapoptotic response to therapy was observed exclusively in wild-type TP53 colorectal cancer models.
It is suggested that pharmacologic and genetic KRAS inhibition differentially remodel the intratumoral T cell compartment, whereas both induce similar T cell changes within the liver premetastatic niche.

This work unveils an unforeseen tumor-suppressing role for mTORC2 in the early adaptation phase of BRAF V600E melanoma cells to targeted therapy and identifies the NAMPT-ETC axis as a potential therapeutic vulnerability of low RICTOR tumors.

Combined BRAF/MEK and PSGL-1 targeting provides a durable anti-tumor response through enhanced functionality and a memory-like phenotype in CD8⁺ T cell subsets.
This study defines adaptive resistance mechanisms to KRAS-targeted therapy in PDAC, revealing both cell-state-dependent RTK rewiring and a conserved SFK adaptive signaling axis and provides a mechanistic rationale for either KRAS/pan-RAS or pan-RAS/SFK co-inhibition as strategies to improve responses to KRAS-targeted t...
