TL;DR

It is demonstrated that PTX3 promotes immune evasion in breast cancer by upregulating PD-L1 by increasing the expression of the E3 ligase RNF216 and caused Beclin-1 degradation, which suppressed autophagy, thereby preventing autophagy-mediated PD-L1 degradation.

Autophagy regulates the function of myeloid-derived suppressor cells in tumor-induced immunosuppression

Tumor-infiltrating MDSCs were isolated from syngeneic C57BL/6 mouse models bearing B16-F10 melanoma or EO771 breast carcinoma tumors. Immunofluorescence staining of LC3B and western blotting of p62/SQSTM1 were used to assess autophagic activity. T-cell suppression by MDSCs was evaluated using co-culture assays, includi...

IL-23 promotes PD-L1 expression and tumor immune evasion via METTL16 pathway.

It is elucidated that IL-23 upregulates METTL16, which in turn enhances PD-L1 expression and facilitates tumor immune escape, which establishes the IL-23/METTL16/PD-L1 axis as a novel regulatory pathway and potential therapeutic target in cancer immunotherapy.

Exonuclease 1 drives immune escape in lung adenocarcinoma by regulating the HIF-1α/PD-L1 axis

Lung Adenocarcinoma (LUAD) represents the most prevalent subtype of lung cancer and additionally possesses the highest potential for lethality. EXO1 is overexpressed in multiple cancers and linked to poor prognosis, but its role in LUAD immune regulation remains unclear. Immunosuppression can be stimulated by HIF-1α, w...

Autocrine Control of CD276 by SPARC in Liver Cancer and Its Contribution to Immune Escape and Resistance to Immunotherapy

ABSTRACT Hepatocellular carcinoma (HCC) frequently demonstrates resistance to immunotherapeutic interventions, a phenomenon largely linked to mechanisms of immune escape. Among the modulators of the tumor microenvironment, SPARC (Secreted Protein Acidic and Rich in Cysteine), a matricellular glycoprotein, has been asso...
